1.Objective:Acute eccentric exercise can induce an increasing level of skeletal muscle hydrogen peroxide(H2O2), it causes oxidative stress and EIMD and the symptoms of DOMS. Studies have proved that acupuncture can speed up the recovery process after muscle damage. The aim of this study is to observe the influence of eccentric exercise on body and the effect of acupuncture or Gd Cl3 on skeletal muscle mitochondrial H2O2 and cytoplasm H2O2 after a bout of eccentric exercise, then investigate the characteristics of acupuncture and the possible mechanism as well as its effect on injury recovery during skeletal muscle damage.2.Methods:The study is divided into two grops, to explore the effect of acupuncture and acupuncture sites, respectively.Group I: 128 healthy rats were randomly divided into four groups: control(C), exercise(E),exercise and acupuncture(EA), exercise and acupuncture impedance(EAI). E、EA、EAI groups rats were exercised simultaneously on a motor-driven treadmill at 16 m min-1down a 16 deg decline continuously for 90 min. EA group use acupuncture intervention(With a diameter of0.25 mm needle sideways pierce quadratus, retaining needle 4 min), EAI use acupuncture intervention and the rats tails intravenous inject Gd Cl3 solution. After the downhill exercise, the exercised rats were subdivided randomly into one of six subgroups, which were killed at6,24,48,72,120 h after the cessation of exercise. Then take the red quadriceps muscle and detect skeletal muscle mitochondrial H2O2, muscle H2O2, cytoplasm H2O2, serum H2O2 concentration and skeletal muscle MDA and SOD value.Group II: 48 healthy rats were randomly divided into four groups: control(C), electrical stimulation(ES), electrical stimulation and acupuncture(ESA), electrical stimulation and acupuncture impedance(ESAI). ES、ESA、ESAI groups rats get a trapezius muscle electrical stimulation eccentric contraction( pulse width 700 ms; 8 v voltage; frequency of 100 hz;electrical stimulation every 3 seconds, 3s × 50 times every set, a total of 10 sets; take five minutes between each group). At the rest time, E group use acupuncture intervention. EAI group use acupuncture intervention and the rats tails intravenous inject Gd Cl3 solution. Immediately after the electrical stimulation take the trapezius muscle and detect cytoplasm H2O2 in vivo or take the trapezius muscle and detect mitochondrial H2O2, muscle H2O2 in vitro.3.Results:3.1 After an acute eccentric exercise, rats bio-chemical criterion in red quadriceps muscle in different groups.Compared with control group, 6~48h after an acute eccentric exercise, rats in E group and EAI group got a significantly rise in mitochondrial H2O2, muscle H2O2, serum H2O2, muscle MDA and muscle SOD(P<0.05). 48~120h post-exercise, rats got a decline in mitochondrial H2O2, muscle H2O2, serum H2O2 and muscle MDA and muscle SOD(P<0.05), and back to normal at the time of 120 h post-exercise.Compared with control group, 6~24h after an acute eccentric exercise, rats in EA group got a significantly rise in mitochondrial H2O2, muscle H2O2, and serum H2O2, mucle SOD(P<0.05).24~120h post-exercise, rats got a significantly decline in mitochondrial H2O2, muscle H2O2(P<0.05), muscle MDA changed little but got a rise at the time of 72 h post-exercise(P<0.05),then back to normal at the time of 120 h post-exercise. SOD kept at a normal level, but got a significantly rise at the time of 72~120h post-exercise.3.2 After an acute electrical stimulation, rats bio-chemical criterion in trapezius in different groups.Compared with control group, after an acute electrical stimulation, Rats in ES, ESA and ESAI group got a significantly rise in mitochondrial H2O2, muscle H2O2 and cytoplasm H2O2(P<0.05). Compared with ES group, rats in ESA group got a significantly decrease in cytoplasm H2O2(P<0.05).4.Conclusion:4.1 An acute eccentric exercise can increase the value of mitochondrial H2O2, cytoplasm H2O2 and muscle H2O2, it induced oxidative stress and reached the highest damage degree at the time of 48 h post-exercise. Then decline and back to normal at the time of 120 post-exercise.4.2 Acupuncture can immediately rise the value of mitochondrial H2O2 and muscle H2O2,decrease the value of cytoplasm H2O2.4.3 Gd Cl3 can obviously decrease the effect of acupuncture, acupuncture effect through the stretch—activated channels(SAC) to get the injury skeletal muscle recovered.4.4 Acupuncture’s effect site is probably located in mitochondria.
