Purposes:To investigate that the effect of hypoxia and hypoxic exercise on Integrin, Focal Adhesion Kinase, Paxillion, the major proteins of Facal Adhesion Complexes of skeletal muscle, the adaptation of hypoxia and hypoxic exercise on Integrin, Focal Adhesion Kinase, Paxillion of skeletal muscle and the different effect of hypoxic exercise on FAK and Paxillion in digitorum longus and soleus.Methods:Male SD rats fell into three parts:chronic hypoxia and hypoxic exercise (12.7% oxygen concentration, train increasing load progressively on treadmill of 10 degree for 4 weeks), hypoxic stress (10% oxygen concentration,7 days), and acute hypoxic exercise (12.7% oxygen concentration, train intermittently on treadmill of 10 degree for 30 minutes, recovery in normoxic condition and 12.7% oxygen concentration for 7 days). Gastrocnemius muscle, extensor digitorum longus and soleus were collected. The content of integrin、FAK、Paxillion of gastrocnemius muscle was determined by Western blotting. The mRNA expression of integrin、FAK、Paxillion was detected by Real-time fluorescence quantitative PCR. The content of FAK, Paxillion and the activity of FAK in digitorum longus and soleus was determined by fluoroimmunoassays histochemical technique (Confocal Laser Scanning Microscopy)。Results:(1) hypoxic stress:there is a tendency of decrease on content of integrin and paxillion in gastrocnemius muscle, but the genetic expression of integrin increases; there is a tendency of descendent followed by increase of the content, the genetic expression and the activity of FAK. (2) After acute hypoxic exercise, the content of integrin, FAK and PAX and the activity of FAK goes up significantly. (P<0.05), and recovers to the level of control quicker under the normoxic condition, compare to hypoxic condition. The influences of acute hypoxic exercise on SOL is larger and earlier than which’s on EDL. (3) Simplex hypoxic stress and simplex exercise can stimulate the synthesis of the protein in Focal Adhesion Complex of skeletal muscle and generate superimposed effect. (4) The content of FAK, Paxillion and the activity of FAK in SOL are much more than in EDL (P<0.05). (5)In the groups of exercise, the proteins in FAC are more or greater, even high significance larger than that in control during 4 weeks exercises (P<0.05)Conclusion:(1) After 10% hypoxic stimulates, the synthesis of protein induced by hypoxia can not make up the degradation of protein caused by hypoxic stimulate. (2)Chronic hypoxia, hypoxic exercise and exercise can evoke the upgrading the content of integrin and paxillion. There is effect of superposition on skeletal muscle by hypoxia and exercise. (2) The content of protein in FAC can be raised by exercise and the capability of anti-damage caused by hypoxia will improved by eligible training. (3) The proteins of FAC might adapt to the hypoxia and hypoxic exercise. After acute hypoxic exercise, they are recovered quickly in normoxic condition and adapt earlier in hypoxic condition after the suitable training. (4)ypoxic exercise effects slow muscle much more than fast muscle and slow muscle can adapt hypoxic conditional better.论文随机验证编号:BT5101717471728...
