Objective:Colon cancer is a common malignancy in China. Surgery remains the only treatment with curative potential for colon cancer. With the further study of the pathogenesis of cancer, targeted therapy has achieved certain curative effect.The aim of this research is to investigate the effect of Transmembrane Tumor necrosis factor-alpha monoclonal antibody in colon cancer in vivo and in vitro.Methods:1. Detect the expression of TM-TNF-a in colon cancer cell lines;2. Detect the effect of TM-TNF-α mAb and chemotherapy drug on the proliferation of colon cancer cell lines;3. Detect the ADCC and CDC effect of TM-TNF-α mAb in colon cancer cell lines;4. Construction of colon cancer model of nude mouse and detection of inhibition of TM-TNF-α mAb on growth of colon cancer.Results:1. TM-TNF-α mAb inhibited the growth of colon cancer cells through ADCC and CDC effect. The expression of TM-TNF-α was higher in colon cancer tissues than that in normal tissues.2. TM-TNF-α mAb inhibited the growth of colon cancer remarkably without extending the survival term.Conclusion:In vitro experiment showed that TM-TNF-a mAb markedly inhibited the proliferation of colon cancer cells through ADCC and CDC effect. In vivo experiment showed that TM-TNF-α mAb significantly inhibited the growth of subcutaneous tumor. TM-TNF-α may be a new target in colon cancer therapy.
