This study observed full-thickness skin defect wound of the back through skin autograft and skin allograft experiment in 3-4 months wuzhishan miniature pig inbred line. Key observation included WZSP survival, the survival of the transplanted skin, local and systemic immune rejectio in 28 days after skin allograft and skin autograft.During the experimental observation of skin allograft and skin autograft in 12 inbred WZSP, the operation process completed successfully including skin graft, sewing, packaging and binding. All inbred WZSP were abnormal in breathing, heart rate and other life indications. Until the experiment end after 28 days, the animal body temperature were normal, normal diet, skin wound healed well, no obvious infection, bad phenomena such as red, dissolved, fall off and necrosis in early wound.In 7 to 14 days after the skin transplantation in two groups, transplantation wound skin ruddy, showed that skin building good blood supply, the skin transplantation survival rate were 100%.Transplant wound had no acute rejection phenomenon such as obvious swelling, seepage, blisters, desquamation, fall off, ulcers, necrosis and so on. IL-4 and IL-2 content in plasma, showed the group after transplantation had no significant increase or decrease before transplantation, there was no significant difference between both. The percentage of CD4+and CD8+in whole blood lymphocytes and the ratio change of CD4+/CD8+, in 14 days after transplantation had decreased, but no statistical differences between two groups, with its dynamic changes were basically identical. Histologic examination had no obvious differences between the two groups. In 7 days after transplantation with two groups, CD4+expression was increased, but there was no significant difference between the two groups.Skin transplantation after 14 days, two groups grafted skin slightly darker, but observed its survival rate were over 80%, and had not o bvious signs of the late rejection such as transplant skin swelling, seepage, blisters, desquamation, dissolving and necrosis and so on. The concentration change of IL-4 and IL-2 in plasma at different time points after transplantation had no significant difference before transplantation. In 21 and 28 days after transplantation, the ratio of CD4+ and CD8+in Whole blood lymphocyte showed reduced than before transplantation, but no statistical difference between two groups. Histology detection had no obvious differences between the two groups. The results showed that the late migration has no obvious immune rejection in the two groups, there was no significant difference between two groups of skin allograft and skin autograft.The results showed that not only skin allograft group but also skin autologous group, there was no immune rejection until 28 days after the experiment.Thus the conclusion:(1)The experiment successfully reproduced skin allograft and skin autograft model of the inbred WZSP, obtained scientific and reliable experimental data.(2) Under the condition of this experiment, skin autograft and skin allograft of the inbred WZSP, has no early and late immune rejection phenomenon.(3) In this experiment, the detection, identification technology of immune rejection had been establishedand through the skin allograft methods of inbred WZSP, accessed scientific and reliable experimental data. It provided a scientific basis of the pig inbred line detection, identification by use of skin grafts technology For the future.
