Miniature pigs had extensively been used in medical and scientific researches in recent years, and the researches on its reasonable anesthesia were more and more. The specificity antagonist for miniature pigs' combined anaesthetic was composed by atipamezole, naloxone, flumazenil and other drug compositions, which was designed for specific combined anesthetic for miniature pigs (XFM), and its recovery process was safe, smooth and fast. Complications of the circulatory system were very common in recovery process after anesthesia, so the effects of specificity antagonist for miniature pigs' combined anaesthetic on hemodynamics and its mechanism in miniature pigs were researched in this study, which was in order to investigate the safety, effectiveness and applicability of the specificity antagonist for miniature pigs' combined anaesthetic.Chinese experimental miniature pigs were injected specificity antagonist for miniature pigs' combined anaesthetic to wake up in three experimental groups (early, middle and late experimental group) after injected XFM in this study. Datex monitor were used to monitor the hemodynamic changes during the period of using specificity antagonist for miniature pigs' combined anaesthetic continuously and dynamicly, simultaneously, blood samples were taken. NO in serum was measured by nitrate reductase, while ET, PGI2, TXA2, PRA, AⅡ, ALD, ANP, CGRP, NPY and NT concentration changes in plasma were mearsured by ELISA. The aim of the study was to investigate the effects of specificity antagonist for miniature pigs' combined anaesthetic on hemodynamics and its mechanism in miniature pigs, which was explained from the endothelium-derived vasoactive substances, R-A-A-S and neural peptides. The results were as follows:After being injected specificity antagonist for miniature pigs' combined anaesthetic 0.08mL·kg-1 in early, middle and late experimental groups, all of the miniature pigs showed a good wake up effect. The recovery process was smooth, and over-exciting, sleeping again, breathing and heart rate too fast didn't appear. The hemodynamic changes during the recovery process were significant and similar in the three groups. The results showed that SBP, DBP, MAP and HR were demonstrated the trend of rising and then decreasing, however, the changes were in range of physiological tolerance. After being injected specificity antagonist for miniature pigs' combined anaesthetic in eraly experimental group, plasma ET, TXA2, NPY, NT, CGRP and hemodynamics changes demonstrated consistent trend, and significant or extremely significant correlations were observed between them. The results showed that ET, TXA2, NPY, NT and CGRP participated in changing of hemodynamics which was caused by specificity antagonist for miniature pigs' combined anaesthetic in miniature pigs anesthetized 10min by XFM.After being injected specificity antagonist for miniature pigs' combined anaesthetic in middle experimental group, plasma PGI2, TXA2, AⅡ, ANP, CGRP and hemodynamics changes demonstrated consistent trend, and significant or extremely significant correlations were observed between them. The results showed that PGI2, TXA2, AⅡ, ANP and CGRP participated in changing of hemodynamics which was caused by specificity antagonist for miniature pigs' combined anaesthetic in miniature pigs anesthetized 30min by XFM.After being injected specificity antagonist for miniature pigs' combined anaesthetic in late experimental group, plasma PGI2, PRA, AⅡ, NT, ANP, CGRP and hemodynamics changes demonstrated consistent trend, and significant or extremely significant correlations were observed between them. The results showed that PGI2, PRA, AⅡ, NT, ANP and CGRP participated in changing of hemodynamics which was caused by specificity antagonist for miniature pigs' combined anaesthetic in miniature pigs anesthetized 60min by XFM.
