ObjectiveCurrently study based show, interleukin 6 (IL-6) gene polymorphism susceptibility genes in a variety of immune-related diseases. Recent studies have shown, IL-6 cells play an important role in respiratory allergic diseases. The study found, IL-6 in the pathogenesis of allergic rhinitis has a clear correlation. To this end, the present study discussed IL-6 gene expression and polymorphisms associated with susceptibility to allergic rhinitis.Methods(1) We have detected histopathological changes in the nasal mucosa of 45 cases of allergic rhinitis patients and 15 cases of nasal septum deviation patients inferior turbinate tissue (control group) by HE staining, and the expression of IL-6 detected by immunohistochemistry.(2) A total of 265 patients with allergic rhinitis and 265 controls were collected in our hospital. Genotyping of IL-6 rs 1800795 and rs 1800796 was performed by polymerase chain reaction (PCR) coupled with restriction fragment length polymorphism (RFLP).(3) We have established a mouse model of allergic ovalbumin using Balb/c mice were divided into six groups according to the excitation time (30min?1h?2h?6h? 12h?24h), while using saline as a control group, each group of five mice. Each group of mice nasal tissue to detect pathological changes in nasal mucosa of mice in each group using HE staining, western blot assay IL-6 expression in the nasal mucosa of mice.Results(1) HE staining showed that, in patients with allergic rhinitis inferior turbinate tissue partially detached, Perspective able to see goblet cell hyperplasia, presence of tissue edema, glands, blood vessels dilate, among a large number of tissue inflammation cell infiltration, including eosinophils, basophils, lymphocytes, neutrophils, eosinophils, which are more common andlymphocytes. Two sections of tissue eosinophils were counted, results showed that allergic rhinitis group eosinophil infiltration significant for (31.92 2.95), normal control group (3.83 2.03), allergic rhinitis group was significantly higher in the control group, both of which had significant difference (P= 0.001<0.05);western blot test results showed that the relative expression of IL-6 protein in patients with allergic rhinitis rhinitis nasal mucosa was (0.40 0.16), whereas in normal controls group turbinate tissue relative expression levels (0.26 0.14), allergic rhinitis group was significantly higher than the control group (P= 0.003<0.05).(2) By x2 test, we found significant differences in the allele frequencies of IL-6 rs 1800795 between patients with allergic rhinitis and controls (x2=4.52, P value=0.03). By multivariate logistic regression analysis, we found that individuals with the C allele of IL-6 rs 1800795 were associated with increased risk of allergic rhinitis when compared with G allele, and the adjusted OR was 1.31 (1.01-1.68).(3) Ethology observations indicate that a mouse model of allergic rhinitis successful modeling; HE staining results showed that each of the nasal mucosa of allergic rhinitis model group there are different degrees of structural disorder, also visible edema and the mucous layer thickening phenomenon, in addition to associated with epithelial tissue necrosis and shedding, ciliated epithelium have fusion phenomenon, deformed nuclei, irregular in shape, columnar cells and goblet cell hyperplasia, vascular dilatation and proliferation, a large number of eosinophils, mast cells, neutrophils, and lymphocytes. Western blot analysis showed that the mice in each group were nasal mucosa IL-6 protein, wherein the relative expression of IL-6 in 30 min group was 0.1420.064, in 1h groups relative expression was 0.2140.104, the relative expression of excitation in 2 h group was 0.2740.092, the relative expression of excitation in 6h group was 0.3240.122, and then over time decreased gradually, in 12h groups relative expression was 0.2360.059, and in 24h group was 0.1720.058. Compared with the control group, the experimental group were all nasal mucosa IL-6 protein expression levels were significantly increased.ConclusionsThe expression of IL-6 increased in patients with allergic rhinitis nasal mucosa and susceptibility polymorphisms were significantly associated with allergic rhinitis of IL-6 gene.
