OBJECTIVE:To evaluate the association of Interleukin-25(IL-25)polymorphisms with persistent allergic rhinitis(PER)due to house dust mite in a Chinese Han population of Jiangsu-Anhui provinces.METHODS and SUBJECTS:This study was based on the hospital-based case-control study,and there were 525 patients(range of age:2-64 years)with mite-sensitized PER and the 611 healthy controls(range of age:2-67 years).Blood was drawn for serum total IgE and allergic-specific IgE(against Dermatophagoides pteronyssinus and Dermatophagoides farina)assay,as well as physical examination,collecting the relevant clinical basic information and genomic DNA extraction.Tagging single nucleotide polymorphisms(tag SNPs)selection were referred to the HapMap data of Chinese Han population in Beijing with minor allele frequency(MAF)?0.05 and r2?0.8,thus finally combine the references three tag SNPs(rs10137082?rs7145551 and rs3811178)in IL-25 were selected for TaqMan genotyping.Logistic regression was performed to analysis the association between selected tag SNPs with PER and using HAPSTAT3.0 software to analyse the haplotype.Generalized Multifactor Dimensionality Reduction(GMDR)was used for gene-gene interaction analysis.RESULTS:The population basic situation analysis indicted no significantly difference in age and gender between the cases and healthy control group(P>0.05).The interquartile range(IQR)of serum total IgE in case and healthy control group were 211.7(97.355-454.735)and 33.20(15.58-62.26),respectively,the case group was higher than control group and the significantly difference was observed(P=0.000).After adjusting for age and gender in Logistic regression,IL-25 polymorphisms were not associated with PER.Subgroup analysis indicated in<25 years group,mutant homozygote(TT genotype)of rs10137082 was related with increased risk of PER,P=0.036,odds ratio(95%confidence interval)were 4.056(1.097-15.001).GMDR analysis show no significantly gene-gene interactions were found with PER by and no significantly haplotype was observed to be with PER.CONCLUSIONS:The genetic variation of IL-25 gene were rs10137082?rs7145551 and rs3811178,which may have no association with PER,while the age may modify the association between rs10137082 loci and PER,further study on signaling pathways would help in understanding of the pathogenesis of AR and identification of targets for clinical therapeutic intervention.
