ObjectiveTo investigate the association of hypospadias and glutathione S transferase enzyme gene GSTT1,GSTM1,GSTP1 and CYP1A1 gene polymorphism.To investigate the mechanism of hypospadias in China.To provide scientific theory for the prevention and treatment of hypospadias.MethodsBlood samples were collected of 120 patients with hypospadias(the case group)and120 normal healthy men(the control group).GSTT1/GSTM1/GSTP1/CYP1A1 genetype was identified by polymerase chain reaction(PCR).The GST activity n the blood serum was detected with Elisa.The content of gonadal hormone in the blood serum of everyone was detected.Analysis of the patients'birth situation and their mothers'epidemiological situation during pregnancy.The data were analyzed with statistical software.The rats of genetype of two groups were analyzed.The interaction betweenthe various facors is analyzed.ReaultsPassive smoking and pesticide/chemical exposure during pregnancy increased the risksofhypospadiasinbirthrate[(?~2=4.676,P<0.05);(?~2=6.330,P<0.05);(?~2=11.997,P<0.01)].The incidence of low birth weight of the hypospadias group 8.3%were high than that of the control group1.7%(?~2=5.614,P<0.05).The GST activity of the hypospadias group(3.89±1.13)ng/ml was lower than that of the control group[(4.232±1.37)ng/ml](t=2.088,P<0.05).The rat of GSTT1(-)Genotype of the hypospadias group59.2%was higher than that of the control group(45.8%)(?~2=4.277,P<0.05).Genotype GSTT1(-)increases the risk of hypospadias risk(OR=1.712,P<0.05).The rat of GSTM1(-)Genotype of the hypospadias group55.8%was higher than that of the control group(38.2%)(?~2=2.003,P<0.01).Genotype GSTM1(-)increases the risk of hypospadias risk(OR=1.925,P<0.05).Passive smoking and pesticide/chemical exposure during pregnancy may be the risk factors for hypospadias.The risk factors of hypospadias can be affected by the joint genotype polymorphism.The rate of hypospadias can be affected by the exposure of risk factors during pregnancy.ConclusionLow birth weight,passive smoking and pesticide/chemical exposure during pregnancy may be the risk factors for hypospadias.There may be closely relationship between GSTT1(-)gene polymorphisms and the incidence of hypospadias.GSTT1 may be the andidate gene of hypospadias.There may be closely relationship between GSTM1(-)gene polymorphisms and the incidence of hypospadias.GSTM1 may be the andidate gene of hypospadias.There may be closely relationship between GSTM1(-)gene polymorphisms andtheincidenceofhypospadias.Thegenetypesof T1(-)/M1(-)/P1(Ile-Ile)/MSP(m1m2,m2m2)andT1(-)/M1(-)/P1(Ile-Val,Val-Val)/MSP(m1m2,m2m2)may increase the rate of hypospadias.The genetype of T1(+)/M1(+)/P1(Ile-Ile)/MSP(m1m1)may decrease the rate of hypospadias.
