Objective:The bone marrow mesenchymal stem cells(BMSCs)of SD rats labeled with iodine-131 ZGO and functionalized self-assembling nano-peptide scaffolds RADA-SKP co-injected into the myocardium of Balb/c mice with acute infarction.The mice were performed multimodal imaging methods including echocardiography,SPECT/CT and PET/CT,to assess changes in cardiac function,to track the localization,survival and migration of BMSCs,and to assess changes in myocardial activity.Combined with pathological findings,to explore the functionalized self-assembled peptide nanoscale scaffolds improving stem cells in treating acute myocardial infarction treatment,and the mechanism.Method:ligation of anterior descending artery induced acute myocardial infraction of Balb/c mice.Group A(blank control group)was treated with 10%sucrose solution;group B(peptide control group)was treated with functionalized self-assembled nano-peptide scaffolds RADA-SKP sucrose solution;group C(cell control group)was treat with endocytic iodine-131 labeled ZGO BMSCs;group D(peptide with cell experimental group)was treat with endocytic iodine-131 labeled ZGO BMSCs combined with functionalized self-assembled nano-peptide scaffolds of RADA-SKP in sucrose solution.Echocardiography assessed cardiac function 1 day before and 1st,7th and 14th day after operation.SPECT/CT was performed on the 1st,6th and 13th day after operation.PET/CT was performed on 2nd and 14th day after operation.The pathology of heart tissue was removed on the 14th day after operation.HE staining and CD34 immunohistochemistry were performed.Results:Cardiac function:The use of functionalized self-assembled nano-peptide scaffolds RADA-SKP can significantly improve the therapeutic effect of BMSCs on the mice model of myocardial infarction.The use of BMSCs alone in the treatment of myocardial infarction model has no significant effect of improve the cardiac function.Stem cell localization:the use of BMSCs combined with the functionalized self-assembled nano-peptide scaffolds compared to only use BMSCs,allows cells to maintain activity for longer periods.The activity of infarcted myocardium:it was significantly improved the infracted myocardium activity by using the functionalized self-assembled nano-peptide scaffolds RADA-SKP combined with BMSCs.At the same time,there was no significant difference in the repair of myocardium in the infarcted area of only use BMSCs compared with co-injection the functionalized self-assembled nano-peptide scaffolds RADA-SKP.Conclusion:The BMSCs injected with endocytic iodine-131 labeled ZGO were injected into the myocardium of Balb/c mice with acute myocardial infarction.The effects of echocardiography,SPECT/CT and PET/CT multimodal examination were used to evaluate the changes of cardiac function,to track the localization,survival and migration of BMSCs,and to assess changes in myocardial activity.BMSCs alone injected into the myocardium of Balb/c mice with acute myocardial infarction could improve the activity of myocardium,but the survival time was short and the improvement of cardiac function was not remarkable.Combined with functionalized self-assembled nano-peptide scaffolds RADA-SKP can prolong the survival time of BMSCs in myocardium,improving the cardiac function and myocardial activity of infarcted area.
