Objective: To investigate the relationship between macrophages and related cytokines and hepatic fibrosis of Echinococcus granulosus,and to investigate the effect of different polarized macrophages on the hepatic fibrosis of mice infected with Echinococcus granulosus influences.Methods: 1)Liver tissue samples and serum samples were collected from 30 patients with hepatic cystic echinococcosis.Masson staining of liver tissue was used to understand the degree of hepatic fibrosis;The expression levels of TGF-?1,IL-17,IL-12 and IL-10 in liver and serum were detected by immunohistochemistry and ELISA respectively;The percentage of CD68/86 + macrophages and CD68/206 + macrophages in peripheral blood of patients with hepatic cystic disease was detected by flow cytometry.2)96 BALB/c female mice were randomly divided into experimental group and control group.The mice in the experimental group were treated with the portal vein larvae to prepare the mouse model of the echinococcus granulosus,and the control group was injected with portal vein of saline;Mice liver tissue,peritoneal lavage fluid and serum samples were collected on day 2,day 8,day 30,day 90,day 180 and day 270 of vaccination;The liver tissue Masson staining was used to study the hepatic fibrosis in mice infected with Echinococcus granulosus;The changes of macrophage F4/80 + CD16/32 + macrophages and F4/80 + CD206 + macrophages in mouse peritoneal lavage fluid were detected by flow cytometry;The expression and localization of TGF-?1,IL-17,iNOS,IL-12,IL-10 and Arg-1 in the liver were detected by immunohistochemistry;The levels of serum TGF-?1,IL-17,iNOS,IL-12,IL-10 and Arg-1 in mice were detected by ELISA;The expression of TGF-?1mRNA,IL-17 m RNA,i NOSmRNA,IL-12 mRNA,IL-10 mRNA and Arg-1mRNA in the liver of mice were detected by real-time fluorescent quantitative RT-PCR.3)Mouse bone marrow-derived macrophages were isolated from culture and induced by macrophages.Macrophages were labeled in vitro for 45 days in mice infected with Echinococcus granulosus.After 90 days of transfusion,mice were harvested for Masson staining,Sirius red staining,liver hydroxyproline assay,Desmin immunohistochemical staining,evaluation of different types of macrophages after transfusion of liver fibrosis changes and treatment.Results: 1)Masson staining of liver tissue in patients with hepatic cystic gland disease showed that patients with different degrees of fibrosis in the live;The levels of TGF-?1,IL-17,IL-12 and IL-10 in liver tissue samples and serum samples were higher than those in normal group;The percentage of CD68/86 + and CD68/206 + macrophages in peripheral blood of patients with cystic hydatid disease was higher than that of normal subjects by flow cytometry.2)mice infected with Echinococcus granulosus: Masson staining of liver tissue showed that with the prolongation of infection time,liver fibrosis and liver fibrosis gradually increased.The results of flow cytometry showed that F4/80 + CD16/32 + macrophages in the peritoneal lavage fluid of mice were higher than those in the control group(P<0.05)at all time points after infection compared with the control group.F4/80 + CD206 + macrophages in the infection 180 days there was a significant increase.The expression of i NOSmRNA and IL-12 mRNA in the early stage of infection was significantly higher than that in the control group(P < 0.05).The expression of IL-10 mRNA and Arg-1mRNA were significantly increased in the early stage of infection.3)M0 and M1 macrophages after transfusion of liver fibrosis than before the return of light.Conclusions: 1)macrophages participate in the process of hepatic fibrosis caused by Echinococcus granulosus infection.2)F4/80 + CD16/32 + macrophages and F4/80 + CD206 + macrophages may be one of the causes of hepatic fibrosis in mice infected with Echinococcus granulosus.3)M0 macrophages and M1 macrophages can reduce the deposition of collagen fibers in mouse liver,whILe M1 macrophages can reduce the collagen fiber deposition in mice.
