Objective 1.To find the certain methods of proteomic analysis in Echinococcus granulosus . 2.To identify some protein by two-dimensional electrophoresis and mass spectrometry.Methods Obtained protein from specimen and quantified the contents of protein. The protein of Echinococcus granulosus or cyst wall was separated by two- dimen sional electrophoresis separating technique, according to different isoelectric points and molecular weight. Used scanner to acquire the picture of 2-DE gel, which was analysed by PDQuest software. The gel was put into cutting machine that would cut and collect dots automatically through choosing the target protein by software.These dots must be enzymolysed by trypsin firstly. And then, they were detected and acquired Peptide Mass Fingerprint (PMF) by MALDI-TOF- MS. By use of Mascot search engine, Compared the PMF with the record of database to identify every protein.Results 1.In 2-DE gels of Echinococcus granulosus and cyst wall,there were 233 and 107 protein dots had been got respectively.2.By using MALDI-TOF-MS,it had 106 and 42 protein dots got the PMF in Echinococcus granulosus and cyst wall.3.The number of identified protein of Echinococcus granulosus was 23 and cyst wall was 14.These protein contained: Actin,Tropomyosin, myosin, Paramyosin, dynactin subunit,Putative peroxiredoxin, Tansketolase, Glycogen synthase kinase-3, Calcium-binding protein,Protein tyrosine kinase, Serine/ threonine-protein kinase,Cytochrome c, ADP/ATP carrier protein ,Phospho- lipase A2, Fatty acid-binding protein, Cyto -chrome P450,Apolipoprotein A-I and Serum albumin.Conclusion 1.The way of proteomic research in Echinococcus granulosus has been found.2.Some proteins have been identified which are involved in the move- ment, growth, development and regulation of the parasite.
