Objective: To study the relationship between variable benign epilepsy of childhood with centro-temporal spikes and the change of the concentrations of zinc and metabolism-associated hormonesMethod: 1.We chose 58 cases of variable benign epilepsy of childhood with centro-temporal spikes as the experimental groups.While 120 children with typical benign epilepsy of childhood with centro-temporal spikes were chosen as the control groups.The concentration of zinc,copper,iron and leptin were measured and analyzed for each of the 178 children.Meanwhile,the erythrocytes of the two groups were also determined with Beckman Coulter STKS automatic blood cells analysis meter.The concentration of metabolism-associated hormones were assessed for each of the 178 children.2.The results of MRS,VEEG and IQ test of the experimental groups were retrospectively analyzed.Results: 1.The concentration of serum iron,zinc and erythrocytes descended in experimental group.Analysis of variance showed statistical significances between the experimental groups children and the control group children(P<0.05).The concentrations of Gh,Cortisol,PTH and TSH descended in experimental group.Analysis of variance showed statistical significances between the experimental groups children and the control group children(P<0.01).The concentrations of ACTH,FT3,FT4 and 25(OH)D ascended in experimental group.Analysis of variance showed statistical significances between the experimental groups children and the control group children(P<0.01).Linear correlation analysis among the nine tested indexes showed that sixteen pairs of indexes exhibited significant positive or negative inter-relationships,accounted for 44% of the total thirty-six couples of indexes.The compare of the concentration of serum copper,apoprotein and leptin showed no statistical significance(P>0.05).2.The differences of the results of MRS,VEEG and IQ test showed statistical significance(P<0.05).Conclusion: The concentration of serum iron,zinc and erythrocytes descended in patients with variable BECT.Meanwhile,The concentration of Gh,Cortisol,PTH and TSH descended in experimental group.The concentration of ACTH,FT3,FT4 and 21(OH)D ascended in experimental group.Hence,studying the relations among this kind of variable benign epilepsy,zinc and metabolism-associated hormones could have a unique significance to the prevention and therapy of this kind of variable benign epilepsy.Objective: To observe the seizure activity and record the survival rate after glutamate-induced seizure model.Method: The different levels of hippocampal cells were added in the 10-day primary cultured hippocampal neurons.24 hours later,we observed morphologic change of cells by contrast phase microscope.We tested the neuron survival rate by using trypanblau staining method and investigated the neuron apoptosis rate by TUNEL technique to evaluate the degree of injury on hippocampal and cerebral cortical neurons.Results: After hippocampal and cerebral cortical neurons were induced by different levers of glutamate for 6 hours,parts of cells became round and swelling with the decurtation or deprivation of the apophysis.24 hours later,most of the swollen cells disintegrated.After testing the neuron survival rate by using trypanblau staining method,glutamate could lower the neuron survival rate significantly.In neuron survival rate,there was a very significant difference in experimental groups with different levels compared with the normal group(P<0.01).In addition,the neuron survival rate decreased along with the increase of the concentration of glutamate.After neurons were induced by different levers of glutamate for 24 hours,TUNEL color showed the shrinkage of the cell surface,the diminution of volume and pycnosis.Different levers of glutamate could all induce neurons apoptosis.Analysis of variance showed statistical significances between the experimental groups and the control group(P<0.05,P<0.01).The neuron apoptotic rate was up-regulated following the increase of the concentration of glutamate(100?200?mol/ L).However,the apoptotic rate decreased along with the increase of the concentration of glutamate(100?200?400?mol/ L).Conclusion: 1.This research has established a glutamate-induced seizure model.successfully,which means glutamate was administered on neurons.The neurons apoptosis rate was high and survivai rate was low after glutamate was administered on neurons.Objective: To investigate the effects of TPEN on the survival of neurons in vitro,to observe the protective effects of TPEN on injured neurons,and to provide new solution to the neuronal injury.Method: TPEN(2.5umol/l)was added into neurons.We induced the neuron apoptosis with the low lever of glutamate.The concentration of caspase-3 of apoptotic neurons was checked by immunocytochemistry techniques,and the effects of TPEN on neurons survival were checked by trypanblau staining method and TUNEL technique.We tested the expressions of Bcl-2 and Zinc transports by using real time PCR.Results: The glutamate(100?mol/ L)could cause the decrease of the neuron survival rate.After TPEN was added into hippocampal and cerebral cortical neurons,the neuron survival rate was significantly up-regulated while the neuron apoptotic rate was strongly down-regulated,analysis of variance showed statistical significances between TPEN and control group(P<0.01,P<0.05).The glutamate(100?mol/ L)could increase the expression of caspase-3.The positive expression rate of caspase-3 in the experimental group was obviously down-regulated and the expressions of Bcl-2,Zn T-1 and Zn T-2 were obviously up-regulated,analysis of variance showed statistical significances between the experimental groups and the control group(P<0.01,P<0.05).Conclusion: After TPEN was added to hippocampal and cerebral cortical neuron,we can find the increase of the neuron survival rate,Bcl-2,Zn T-1 and Zn T-2 with the decrease of the neuron apoptotic rate and caspase-3 concentration.TPEN can alleviate neuron apoptosis and improve cell survival by decreasing caspase-3 and increasing Bcl-2,Zn T-1 and Zn T-2 expression.
