Study On The Function Of Shao Yao Gan Cao Decoction Treating Experimental Autoimmune Uveitis Under The Guidance Of Translational Medicine

Objects:1.The deep meaning of 'liver open into eye,treating eye diseases from liver'is explored based on literature research.The liver and eye have the characteristic of embodying yin and function yang.Liver yin deficiency leading fire flaming up is the potential pathogenesis of uveitis.Heat syndrome of eyes is the outside manifestations,liver yin deficiency is the root.Tonifying liver yin is the basic therapeutic method for uveitis.Shao Yao Gan Cao Decoction is the basic prescription of treating uveitis.2.We observe improvement of the signs,symptoms,pathological injury,expression dose of IFN-??IL-10?IL-17 and balance of Thl/Th2 in serum and splenic lymph nodes,balance of Thl/Th2 of EAU rat eyes by using Shao Yao Gan Cao Decoction with different matching.We explore the deep functional mechanism of Shao Yao Gan Cao Decoction treating uveitis which will provide foundation for clinical medication.Methods:1.Theoretical research:We make stasis and analysis on all the prescriptions which have medicine Shao Yao,Bai Shao,Chao Bai Shao,Chi Shao recorded in ancient literatures about eye diseases in Chinese Medical Classics by using stasis software.We analyze the ratio of Shao Yao and Gan Cao in prescriptions by using frequency analysis,association rules analysis and entropy methods.We also use the SPSS Statistics 17.0 software to analyze the functioning meridians of specific medicine to explore the diagnosis and therapeutic principle of cataract eye diseases and other eye diseases and application of Shao Yao and Gan Cao.We also analyze the prescriptions have the medicine Shao Yao and Gan Cao,Chi Shao and Gan Cao recorded in modern literatures.2.Experimental research:We construct EAU rat model by hypodermic injecting IRBP-R16,CFA and mycobacterium tuberculosis for 3 times.We give Shao Yao Gan CaoDecoction with the ratios Shao Yao and Gan Cao of 1:1?2:1?3:1 to rat models by intragastric administration.We observe the inflammatory manifestations of rat models by using Digital slit lamp dynamically.we also make anterior segment photography.We record changes of outside manifestations of eyes after 7,14 and 21 days of model construction.We analyze the data by using Caspi standard score.We make pathological observations,HE stain after intragastric administration.We observe morphological changes and inflammatory cell infiltration.We also test expression dose of IFN-??IL-10?IL-17 and balance of Th1/Th2 in serum and splenic lymph nodes,balance of Th1/Th2 of EAU rat eyes.Results:1.It shows that medicines applied in treating eye diseases mainly function on liver meridians,spleen and heart meridians are as follows.It testified that liver open into eyes and we should treat eye diseases from liver.It shows that medicines used in eye diseases take sweet,bitter,pungent and sour as the main taste.2.Modem literatures research 40 passages are adopted.Baishao occurs 4 times.Chi Shao occurs 32 times.Bai Shao and Chi Shao occur 4 times together.It shows that excess syndrome of the liver meridian is the main consideration.And clear liver fire is the main principle.The latent pathogenesis isn't taken into consideration.3.Influences on the outside manifestations of EAU rat eye of Shao Yao Gan Cao Decoction with different ratios.There are little differences between experimental groups and model group after 1 week of administration.The rats in experimental group improve markedly compared with the rats in model group after 2 weeks of administration.Ciliary congestion disappears.The anterior chamber is normal.Aqueous humor is turbid slightly.The iris is normal.The pupil area disappeared.Fundus red light reflection disappears.The grade in the third group is nearly the same with the blank group.4.Influence on the Pathological Changes of EAU Rats of Shao Yao Gan Cao Decoction with Different Ratios model group,The structure of the retina is disordered and tortuous.it shows cell proliferation,cell vacuolar degeneration.The cell spacing is mildly different.Inflammatory cell infiltrate at each level.Ganglion cell layer,the reticular layer still takes on inflammatory cell infiltration in the first three groups.The symptoms mentioned above alleviate slightly in the second and third group.There is scattered neutrophil infiltration.There are cell degeneration partially.Cell spacing is different.The blood vessels have mild hyperplasia in the first group.5.Influence on the expression of IFN-??IL-10?IL-17 and the Th1/Th2 of the EAU rats of Shao Yao Gan Cao Decoction with different rations.Intragastric administration showed rat serum and lymphocyte factor detection after 2 weeks after intragastric administration of Shaoyaogancao decoction treatment,inflammatory factor IFN-? and IL-17 levels were decreased,inhibiting inflammatory factor IL-10 levels rise,different degree recovery ratio of Th1/Th2 level.Conclusions:1.Statistical results of the ancient literatures shows that the liver meridian is the main therapeutic part of eye diseases.The medicine with sweet,bitter,pungent and sour is the main choose of doctors.Dang Gui and Bai Shao are high frequency used medicine.These two medicine has the function of nourishing liver yin and blood.2.Liver opens into eye.The liver has the characteristic of maintaining the qi flow free.Liver fire grows upwards and consumes the yin-fluid and the eyes lose the nourishment.The study shows that liver yin deficiency and fire flaming upwards is the latent pathogenesis.The heat syndrome of eye is the outside manifestation.The liver lose the nourishment is the root reason.The therapeutic principle should be nourishing the liver.The Shao Yao Gan Cao Decoction is recorded in the book Shang Han Lun which has the function of nourishing liver.This prescription should be used to treat the pupil diseases.3.The literature research provides the evidences for the ratio of Shao Yao and Gan Cao.The most commonly used ratio is 1:1?2:1?3:1 which provides evidences for the experimental research.4.There are big improvements on inflammation and pathological scores of eyes of EAU rats.The Shao Yao Gan Cao Decoction has the function of inhabiting the EAU rats.The ration of 3:1 shows the best effect.5.The possible mechanism of Shao Yao Gan Cao Decoction treating uveitis is acting on the disordered immune function.It will inhabit the proinflammatory factors IFN-??IL-17,increase the expression of antagonistic inflammatory factors IL-10,maintain the balance of Th1/Th2 and maintain the balance of yin and yang.
