Camellia oleifera is widely distributed in the hilly areas of Southern China,and the development of C.oleifera industry is of great significance.But with the continuous development of the C.oleifera industry,the planting area of C.oleifera is keeping expanding and the demand for seedlings is growing.Influenced by the rootstock,spikelet and seedling cycle,the quality seedlings of C.oleifera have been unable to meet the demand of the market.In order to explore ways to accelerate the growth of seedlings and improve the efficiency of seedling raising,19 photosynthesis related genes were cloned,effects of different LED light quality,light intensity and illumination time on the growth and photosynthesis related genes expressions of C.oleifera seedlings were studied,LED light condition suitable for C.oleifera seedlings cultivation and 9 differentially expressed genes related to photosynthesis of C.oleifera were screened out,taking two years old cuttage seedlings of‘Changlin-4'C.oleifera superior clones as test materials.The main results in the paper are as followings:?1?CoRbcS1,CoRbcS2,CopsbA,and so on 19 photosynthesis related genes were cloned with specific primers,taking cuttage seedlings of‘Changlin-4'C.oleifera as materials,using the existing transcriptome data of research group and NCBI database,and uploaded to GenBank?MG746996MG747014?.All of the 19 gene sequences contain complete open reading frames,and the encoding proteins do not contain Pyl and Sec.Of the 19 proteins,there are 13 hydrophilic proteins,6 hydrophobic proteins,14 stable proteins,and 5 unstable proteins.The cloning and analysis of 19 genes provide a basis for the next studying of the genes expressions treated with LED light.?2?The C.oleifera seedlings were treated with 7 different light quality?red LED,90%red and 10%blue,70%red and 30%blue,50%red and 50%blue,30%red and 70%blue,10%red and 90%blue,and blue LED?,taking white LED as control.The research found that the seedlings of‘Changlin-4'C.oleifera treated with the composite light of 10%red and 90%blue had the largest diameter growth,dry matter content,sound seedling index,AQY,Pmax,Gs,Tr,?PS?,ETR,photosynthesis pigment content,ZR content and POD activity,and as well as the highest gene expression of CoFBP,Cocab7,CoTPI and CoTkt.It illustrates that the composite light of 10%red and 90%blue is the ideal light quality for‘Changlin-4'C.oleifera seedlings cultivation.?3?The C.oleifera seedlings were treated with 5 different light intensity(100?mol·m-2·s-1,150?mol·m-2·s-1,200?mol·m-2·s-1,250?mol·m-2·s-1 and 300?mol·m-2·s-1)of 10%red and 90%blue composite light,basing on the screening of the most suitable LED light quality.The study showed that the seedlings of‘Changlin-4'C.oleifera treated with the light intensity of250?mol·m-2·s-1 had the largest seedling height growth,diameter growth,dry matter content,sound seedling index,AQY,Pmax,Gs,Tr,?PS?,ETR,qP,soluble protein content,soluble sugar content,free amino acid content,IAA content,GA content and CAT activity,and as well as the highest gene expression of CorbcL,CoRbcS1,CoRbcS2,CoRCA,CoFBA1,CoSBP,CopsaA,CopsaB,CopsbA,CoTPI,Cocab,Cocab6,Cocab7 and Cocab8.It illustrates that250?mol·m-2·s-11 of 10%red and 90%blue composite light is the ideal light intensity for‘Changlin-4'C.oleifera seedlings cultivation.?4?The C.oleifera seedlings were treated with 3 different illumination time(12h·d-1,14h·d-1 and 16h·d-1)of 250?mol·m-2·s-1 10%red and 90%blue composite light,basing on the screening of the most suitable LED light quality and light intensity.The results suggested that the seedlings of‘Changlin-4'C.oleifera treated with the illumination time of 16h·d-1 had the largest diameter growth,dry matter content,sound seedling index,AQY,Pmax,Gs,?PS?,ETR,qP,soluble sugar content,free amino acid content,SOD and POD activity,and as well as the highest expression of 19 photosynthesis related genes.It illustrates that 16h·d-11 of250?mol·m-2·s-11 10%red and 90%blue composite light is the ideal light condition for‘Changlin-4'C.oleifera seedlings cultivation.?5?Genes of CorbcL,CoRbcS1,CoRbcS2,Co FBA1,CoFBP,CoTkt,CoSBP,CoTPI,Cocab6 and Cocab7 showed significant or extremely significant differences under the treatments of different LED light quality,light intensity and illumination time,it illustrates that these genes are greatly influenced by the LED light source and have important research significance.
