Duck viral hepatitis is a highly fatal contagious disease of ducklings,it can be caused by duck hepatitis A virus(DHAV)and duck astrovirus(DAstV),and DHAV-3 is most prevalent in duck industry in China.Earlier work suggested the possibility to anti DHAV-3 using the resistance breeding strategy.Here,we report the breeding of the second and third generations of Pekin duck resisitance to DHAV-3.Moreover,a highiy resistant flock and a highly susceptible Pekin duck flock were selected to investigate their responses to DHAV-3.Differentially expressed genes were screened from livers of susceptible ducks and resistant ducks using RNA-seq.The study may provide the research materials and cules for futher research on the resistance to DHAV-3.The main conclusions are as follows:1.The third generation of Z8 line exhibits strong resistance to DHAV-3 after three generation breeding.The resistance of one generation to DHAV-3 has been transmitted to next generation.2.DHAV-3 can be replicated rapidly and efficiently in the liver of susceptible flock,whereas the replication of the virus in the liver of resistant flock is suppressed greatly.3.Susceptible flock exhibit a much stronger inflammatory response than resistant flock,resulting in the death of susceptible flock.4.A total of 1490 differentially expressed genes were screened from livers of susceptible ducks and resistant ducks infected with DHAV-3 using RNA-seq.The up-regulate gene which associated with immune response,and down-regulated genes which are related with growth and metabolism in susceptible ducks.5.A total of 75 differentially expressed genes were screened from livers of susceptible ducks and resistant ducks before infection using RNA-seq.The up-regulate gene is mainly related with immunoglobulin and antiviral activity in resisitant ducks,which is the mian reason for them resistant to DHAV-3.6.A total of 28 potential candidate resistant genes were gained.
