Heart Yang deficiency syndrome is one of the common syndromes of cardiovascular system disease,and Heart-Yang deficiency is the main pathological basis of coronary heart disease.Recent studies show that,application of metabonomics technique can reveal the law of development of TCM syndromes and syndrome diagnosis,the essence and basic biology.Therefore,this study was based on 264 cases of large clinical casesthe,Heart Yang deficiency patients as the research object,with treatment by warming yang supplementing heart,with the application of metabonomics technology,to reveal the Heart Yang deficiency syndrome and Heart Yang deficiency syndrome of coronary heart disease(CHD)of Urine metabolomic biomarkers.To provide scientific basis for clarifying the function target of Heart Yang deficiency syndrome and Heart Yang deficiency syndrome of coronary heart disease(CHD)from the perspective of metabolic syndrome and to reveal the biological basis of Heart Yang deficiency syndrome of coronary heart disease(CHD)treated with the method of warming yang to strengthen the heart yang deficiency syndrome.Objective:Study on the urine metabolic biomarkers of Heart Yang deficiency syndrome and Heart Yang deficiency syndrome of coronary heart disease(CHD).To make clear the clinical efficacy and metabolomics mechanism of Heart Yang deficiency syndrome of coronary heart disease(CHD)with the method of warming yang supplementing heart.Clarify the biological basis of heart Yang deficiency syndrome and Heart Yang deficiency syndrome of coronary heart disease.Methods:We used the UPLC-Q-TOF-HDMS technology and data patten recognizing methods(PCA,OPLS-DA)to analyze the urine metabonomics biomarkers of 164 hear t disease patients who were diganosed as Heart Yang deficiency syndrome and 100 he althy volunteers.We chose 87 patients who were also dignosed as coronary heart dise ase to compare their urine metabonomics biomarkers with the 100 healthy volunteers.Furthermore,we treat the 30 coronary-heart-disease patients by the treatment of warm ing yang supplementing heart to observe the clinical effect and analyze the influences to expression of the identified urine metabonomics biomarkers of the heart Yang defic iency syndrome of coronary heart disease.The urine sample was analyzed by UPLC-Q-TOF-HDMS.All data were applied to MassLynx4.1 and data for peak recognition,peak correction and normalization.Importing multivariate data matrix into EZinfo software for pattern recognition and analysis.Data processing is carried out by principal component analysis(PCA)and orthogonal partial least square discriminant analysis(OPLS-DA).Using MetPA software to search for metabolic pathway related to markers.Results:?There were identificated 43 biomarkers of heart yang deficiency syndrome.Such as Citric acid,Uridine,N-Acetyl-L-aspartic acid,Gulonic acid,Citric acid,Hippuric acid and so on.lt was confirmed that the metabolic pathway of heart yang deficiency syndrome was related to citric acid cycle,the metabolism of sugar,the metabolism of pyruvate,and purine metabolism and so on.?There were identificated 31 biomarkers of heart yang deficiency syndrome of coronary heart disease(CHD).Such as Citric acid,Asymmetric dimethylarginine,Uridine,Alpha-LactoseMalonic semialdehyde,2-Furoic acid and so on.It was confirmed that the metabolic pathway of heart yang deficiency syndrome of coronary heart disease(CHD)was related to lemon acid cycle,beta alanine metabolism,propanoate metabolism and pentose and glucuronate conversion and so on.?The warming yang supplementing heart method can improve heart yang deficiency syndrome of coronary heart disease(CHD)in patients with clinical symptoms,improve myocardial ischemia,improve nitroglycerin stop rate.?The Heart Yang deficiency syndrome of coronary heart disease patients were treated by warming yang supplementing heart,to validate 31 differential metabolites which has been identified.Compared before and after treatment,there were7 metabolites content changes,Uric acid,Butyrylcarnitine,IDP,Alpha-Lactose,Citric acid,Uridine,Asymmetric dimethylarginine.The warming yang supplementing heart method can through the intervention of heart yang deficiency syndrome of coronary heart disease(CHD)of urine metabolic biomarkers to improve the clinical effect of the track change.Conclusion:?Determination of urinary metabolic biomarkers in the heart yang deficiency syndrome and heart yang deficiency syndrome of coronary heart disease(CHD),and provide a new basis for differentiation of disease from the perspective of metabolic syndrome in the combination of disease and syndrome.?The differences of the heart yang deficiency syndrome and heart yang deficiency syndrome of coronary heart disease(CHD)biomarkers and metabolic pathway indicate that urine metabolism biomarkers have its unique characteristics in the same syndrome and different disease mode.?By the method of the method of warming yang supplementing heart through callback the Metabolic locus of heart yang deficiency syndrome of coronary heart disease(CHD)to play a role in treatment.It was further confirmed that Uric acid,Butyrylcarnitine,IDP,Asymmetric dimethylarginine,Alpha-Lactose,Citric acid and Uridine were biomarkers of urine metabolism of heart yang deficiency syndrome of coronary heart disease(CHD).
