Platycladi Cacumen,the typical used Chinese medicine,is the dry twig and leaf of Platycladus oriental is(L.)Franco.It has performed as the key medicine for blood-cooling,hemostasis and hair-regrowing.Additionally,it has good effect in hemostatic,the effect is better after processing than that of before processing,few literatures to report the mechanism about this activity until now.So,the study on the processing mechanisms of Platycladi Cacumen and Cacumen Platycladi(carbonized)is considered the key to enhancement of it’s clinic effects.The study of this paper includes the following sections:1.Literature reviewWe have reviewed relative literatures,collate many research results and discussion,and summarize a review about chemical compositions,pharmacological,the effect on processing,quality evaluation,and clinical applications of Platycladi Cacumen,with a view to provide more comprehensive basis to do further research.2.Drying researchEstablish the degradation kinetc model of quercitrin by using chemical kinetics equations with the experiment data.During the dring process,the degradation rate of quercitrin changed with the temperature,time and moisture content was generally considered to follow the law of chemistry kinntic.The degradation of quercitrin under 70℃ water bath was follow:lnQ=-0.003810t+0.2009,R2=0.9764,the rate constant of reaction is 0.0038hr-1,t1/2 was 181.87hr.We establish the degradation dynamic model of quercitrin and get model parameter under 30℃<t<90℃,5%d.b.<M<190%d.b.:K0=30.25hr-1,a1=15385.48KJ/mol,a2=2.17KJ/mol.We get the equilibrium moisture content model of twig and leaf of Platycladus orientalis(L.)Franco through static method,the fitting degree of experimental data and Hednerson model was the best.The model indicate that in certain relative humidity,equilibrium moisture content was inversely proportional to temperature changes;in certain temperature,equilibrium moisture content increases with an increases of the relative temperature.Through the exprement of different moisture content,we draw the curve between lnMR and t and obtain the water diffusion coefficient under different conditions through k.The data had been regressed by using software and the model of water diffusion coefficient of Platycladi Cacumen had set up.It turns out that the water diffusion coefficient is higher when the tempreture or the initial water ratio are higher.3.Processing procedureTaking quercetin as the investigated index,we use intelligent stir-frying machine of drum type to screen the best processing technology by orthogonal test.The result implies that there is huge difference between laboratory and factory.The best parameters is 250℃,processing 6min.The technology is steady,we take it for pilot test to produce 10 batches of samples.The yield rate is higher than 85%.4.Study on the effective part of hemostasia about cacumen platycladi(carbonized)We choose the BT and CT as indexs,do the typical pharmacological experiment and several indexs of clotting system,fibrinolytic system,and platelet aggregation,the result suggested that the flavonoids part is the effective part of hemostasia,which were mediated through shorten PT,APTT,and increase FIB quantity.So we infer that the change of content and component of flavonoids part is closely related to the better effects in hemostatic.However,the impact on the blood system of the test materials was detected by Multi-index,the tannin part also played a role on platelet aggregation.5.Using a multivariate statistical approach to analysis the change of compounds in Platycladi Cacumen before and after processTo obtain the complete profile from a sample,it is necessery to run the LC/MS analysis in both positive and negative modes.The LC/MS dataset was to convert the 3DLC/MS data into a 2D matrix by using the Markerlynx.Markerlynx converts each data point into an Exact Mass Retention Time(EMRT)pair and tabulates the result into a 2D matrix.The data is processed first by using principal component analysis(PCA).Then a minimum supervised statistical model,Orthogonal Partial Least Squaer-Data Analysis(OPLS-DA),can be applied for orthognol data analysis and a list of top contributing markers for each sample can be got.At last,20markers were got for each samle.According to the information of compounds seven markers were identified,which were myricetin3-O-rhamnoside,kaempferol3-O-rhamnoside,myricetin,quercitrin,quercetin,kaempferol and amentoflavone.The main change is that after processing quercitrin transformed to quercetin,it is also verified flavonoid glycosides draw off glycosyl is the main change in Platycladi Cacumen before and after process.6.Uptake determination of quercitrin and quercetin in Caco-2 cells monolayerTo study the absorption of quercitrin and quercetin,Caco-2 cell monolayer was employed to investigate the effect of incubation time.The result shows that the absorption of quercetin is most fast than quercitrin,the cell permeability become better when flavonoid glycosides draw off glycosyl,So we infer that the process of change is closely related to the better effects in hemostatic.7.Study on the effective components of hemostasia about cacumen platycladi(carbonized)We choose the BT and CT as indexs,do the typical pharmacological experiment and several indexs of clotting system,fibrinolytic system,and platelet aggregation,the result suggested that quercetin is the effective part of hemostasia,which were mediated through shorten PT,APTTand increase FIB quantity.Quercetin also reduced the platelet aggregation of induced by ADP.The impact on the blood system of the test materials was detected by Multi-index,though we find that Quercetin made a significant contribution to wound-healing activity of cacumen platycladi(carbonized).
