The purpose of this trial is to examine whether the traditional medicinal materials-Platycladi Cacumen have α-glucosidase inhibitory activity, and to determine the effective parts,then optimize the extraction and separation technology of this part.These are the foundation work for develop Platycladi Cacumen as a new antidiabetic drug,.The test is divided into two sections:Frist Divide the Platycladi Cacumen extract into four parts by using solvent extraction, then assay the a-glucosidase inhibitory activity of each part. We find that the ethyl acetate part have highest activity. Using low-voltage column chromatography,we analyse compositions of ethyl acetate part. And we get two compounds-amentoflavone and quercetin.. Then we assay the α-glucosidase inhibitory activity Amentoflavone have the highter activiy.Comparing the activity and the flavonoid content data of each part, we find that they are positively related. The flavonoids ingredients are the main effective ingredients of α-glucosidase inhibition in Platycladi CacumenSecend Using the flavonoids content as index, we chose the optimization extraction and separation technology. We assay the flavonoids content by the ultraviolet spectrophotometry and choosing amentoflavone as reference substance. The best technology:with70%ethanol extraction Platycladi Cacumen, extract through the AB-8macroporous resin column, add water dilute the effluent, then through the polyamide resin column, throw the outflow away, use80%ethanol to wash down the polyamide resin column, collect the eluent, concentration, to become the Platycladi Cacumen flavonoid.The method is simple,environmental and applying to industrial production.
