In allusion to the treatment of the male bald in clinical medicines,most of them have adverse reactions such as sexual hypoactivity,impotence,irritability.We choosed the drug inhibition of alopecia which avoided the above symptom in the aspect of traditional Chinese medicine.In this study,We choosed compounds which can inhibit 5a-R,making it foam aerosol and researched it on the quality.The main work was as follows:Firstly,the influence factors of extracting Cacumen Platycladi were studied through orthogonal array experiment.The in vitro cytotoxicity of finasteride and ethyl acetate extracts of Cacumen Platycladi against a breast cancer cell line(RM-1)were performed using MTT assay.High Performance Liquid Chromatography method were used to establish 5a-R screening model in vitro.The results showed that total Rutin was 8.28%used the optimal procedures which extracted twice by used 60%aqueous ethanol(V/N)as the extraction solvent with a material/liquid ratio of 1:10 for 1h and used ethyl acetate extraction.This kinds of flavonoids can significantly inhibit the proliferation of RM-1 cell in dose dependent manner.When cells had a higher survival rate,both finasteride and of acetate extracts of Cacumen Platycladi of O.lmg/mL were optimal concentration.High Performance Liquid Chromatography method were used to establish 5a-R screening model in vitro:0.5mL pH6.5 sodium phosphate buffer,O.1mL 50%ethanol,150 μL 0.5mg/mL testosterone,250μL 2.0mg/mL NADPH,0.5mL 5mg/mL enzyme extract,37℃for 30min.The final formulation of flavonoids compounds is Cacumen Platycladi extracting by ethyl acetate which inhibition rate was 73.36±3.53%.The best formulation and preparation of foam aerosol was determined by the performance of bubbles in vitro as evaluation by using single factor test:12%Tween-80,6%Propylene glycol and 81%water.The quality of foam aerosol of Cacumen Platycladi extracting by ethyl acetate was evaluated with the transdermal absorption.The transdermal performance was conducted with the modified Franz diffusion cell and in vitro mice abdominal skin whose specific parameters were permeation volume and transdermal retention volume.The transdermal effect of butenafine hydrochloride nanoemulsion was tested in mouse skin in vitro,with comparation of accumulated transdermal quantity and retention volume among Cacumen Platycladi extracting by ethyl acetate foam aerosol.The permeation volumes of these 2 different drugs were 47.08μg/cm2,45.92μg/cm2,respectively and transdermal retention volumes were 8.24±0.11μg/cm2,8.49±0.67μg/cm2,respectively.So foam aerosol,as a new kind of transdermal drug delivery system,was the same as cream.Studying this subject will provide new way for the treatment of inhibiting bald hair,drug releasing was more stable,reducing costs,suitable for industrial production,and better suitable for the patient’s medication.Lay the foundation for enriching Cacumen Platycladi dosage forms and developing new drug.
