Objective:To observe the effects of Yangxin Decoction on vascular endothelial injury and RhoA/Rockl signaling pathway in rats,and to explore the protective mechanism of Yangxin Decoction on vascular endothelial injury.Methods:50 male SD rats were randomly divided.into blank group,model group,folic acid group,Yangxin Decoction group and fasudil group,with 10 rats in each group.The blank group was given ordinary feed,the other 4 groups were given 3%L-methionine feed,and the vascular endothelial injury model of hyperhomocysteinemia was replicated,and the time of replicating of the model was 12 weeks.Thirteenth weeks,the rats were fed with conventional feed,and the rats in the model group and the blank group were given the conventional equal volume distilled water.The folic acid group was given the folic acid 1.8mg/kg/d gavage;the Yangxin Decoction group was given the nourishing cream of Yangxin soup 2.34g/kg/d,and the fasudi group was given the intraperitoneal injection of fuhudil 2.7mg/kg/d.The drug was given for 4 weeks.At the end of the sixteenth week,the rats were weighed and weighed,and the vascular endothelial cells of thoracic aorta were observed by HE staining.The ultrastructural changes of thoracic aorta endothelium in the thoracic aorta were observed by transmission electron microscope,and the levels of serum NO,ET-1,Hey,EPCR and TM were detected by ELISA,and the vasodilatation of thoracic aorta of rats was detected by blood vessel ring in vitro;The expression of Bax and Bcl-2 protein in thoracic aorta endothelium of rats was observed by immunohistochemical method;expression of RhoA,Rockl and eNOS mRNA in thoracic aorta of rats was detected by RT-PCR;Western Blot method was used to detect the expression of RhoA,Rockl,p-MYPT1,and protein in thoracic aorta of rats.Results:(1)electron microscopy and pathomorphology showed that Yangxin Decoction could alleviate the pathological damage of vascular endothelium and protect the vascular endothelium in rats.(2)Yangxin Decoction can increase the level of serum NO in rats and reduce the level of ET-1,Hey,EPCR and TM(P<0.01),and the effect is equivalent to folic acid and fasudil(P>0.05).(3)Yangxin Decoction could improve the endothelium dependent vasodilatation function of thoracic aorta in rats.The difference of ACh terminal concentration(10-8,10-7,10-6,10-5)mol/L was statistically significant(P<0.05),and the effect was equivalent to folic acid and fasudil(P>0.05),and there was no significant difference between the non endodermis dependent vasodilatation function of each group(P>0.05).(4)the effect of Yangxin Decoction on Bax expression of thoracic aorta and up regulation of Bcl-2 expression in rats was statistically significant(P<0.01),and the effect was equivalent to folic acid and fasudil(P>0.05).At the same time,it could down regulate the protein and gene expression of RhoA/Rockl signaling pathway and up regulate the expression of eNOS protein and gene,and the difference was statistically significant(P<0.01).The effect was similar to that of fasudil(P>0.05).Conclusion:1.Yangxin Decoction can reduce the level of serum ET-1,Hcy,EPCR and TM,increase the level of serum NO,and improve the pathological injury of vascular endothelium of thoracic aorta of the rat model of vascular endothelial injury.2.Yangxin Decoction can improve the endothelium-dependent vasodilation function of isolated thoracic aorta rings in rats with vascular endothelial injury.3.Yangxin Decoction can reduce the expression of Bax in the thoracic aorta of rats with vascular endothelial injury,up regulate the expression of Bcl-2,and inhibit the apoptosis of vascular endothelial cells.4.Yangxin Decoction can reduce the expression of RhoA,RocklmRNA and protein in the thoracic aorta of the rat model of vascular endothelial injury,inhibit the RhoA/Rockl signaling pathway and increase the expression of eNOSmRNA and protein,which may be one of the mechanisms of protecting vascular endothelial injury against AS.
