Objective:To investigate the impact of yangxin decoction on rat with chronic heart failure from Ca2+-CaN-NFAT3 signal pathway.Explore the yangxin decoction treatment of chronic heart failure rat ventricular remodeling mechanism,for yangxin soup will provide the basis for further application in clinical.Methods:90 rats of clean grade male Wistar were selected as normal control group with table of random number.Another 20 rats were selected as yangxin decoction prevention group.yangxin decoction prevention group rats were model replication at the same time to give the Yangxin decoction.The remaining 60 rats were established the rat model of chronic heart model with intraperitoneal injection of adriamycin joint high-fat feed.The successful model of rats were divided into three groups:model control group,yangxin decoction treatment group,captopril treatment group.Every day rats were given drug according to body surface area method,the conversion of drugs are as follows:yangxin decoction treatment group and yangxin decoction prevention group were respectively given 2.3 4g/kg,captopril treatment group was given 2.25mg/kg,besides,the model group and normal group were given equally volume of physiological saline.Every rat was given only once a day for 4 weeks.After the experiment we observed the general condition of rats,the echocardiography index,the concentration of hemodynamics,the changes of myocardial tissue pathological structure,the myocardial ultrastructure,mearsured the concentration of myocardial intracellular calciumion,the activity of CaN in myocardial tissue,protein expression of NFAT3 and GATA4 in myocardial tissue.Result:1.General condition:The model of chronic heart model rats with intraperitoneal injection of adriamycin joint high-fat feed were showed diffict breathing,slow response,dry hair,pale nose,abdominal ascites,stool loose,etc.2.Echocardiographic result:After model copy,compared with the normal group,LVEF of model control group was lower,LVIDs、LVIDd were higher.After 4 weeks,compared with the model group,LVEF of yangxin decoction treatment group,captopril treatment group and yangxin decoction prevention group were higher(P<0.05),LVIDs、LVIDd were lower(P<0.05).3.Hemodynamic result:Compared with normal control group,LVSP、+dp/dtmax、-dp/dtmax of the model control group were lower,LVEDP was higher(P<0.05);compared with the model group,LVSP、+dp/dtma、-dp/dtmaxof the yangxin decoction treatment group,captopril treatment group and yangxin decoction prevention group were higher,LVEDP was lower(P<0.05).4.Results of morphology and ultrastructure of myocardial tissue:Compared with the model control group,the damage of the myocardial cell of the treatment groups were alleviated;the infiltration of inflammatory cell reducted and edema lightened.The myocardial ultrastructure was also significantly improved compared with the model control group.5.The concentration of calciumion result:Compared with normal control group,the concentration of calciumion in myocardial tissue of the model control group was higher(P<0.05);compared with the model group,the concentration of calciumion in myocardial tissue of the yangxin decoction treatment group,captopril treatment group and yangxin decoction prevention groupwere lower(P<0.05).6.CaN content result:Compared with normal control group,the concentration of CaN in myocardial tissue of the model control group was higher(P<0.05);compared with the model group,the concentration of CaN in myocardial tissue of the yangxin decoction treatment group,captopril treatment group and yangxin decoction prevention groupwere lower(P<0.05).7.Realtime PCR result:Compared with normal control group,the expression of NFAT3 mRNA、GATA4 mRNA of the model control group was higher(P<0.05);compared with the model group,the expression of NFAT3 mRNA、GATA4 mRNA of the yangxin decoction treatment group,captopril treatment group and yangxin decoction prevention groupwere lower(P<0.05).8.Western Blotting result:Compared with normal control group,the protein expression NFAT3、GATA4 in myocardial tissue of the model control group was higher(P<0.05);compared with the model group,the protein expression of NFAT3、GATA4 in myocardial tissue of the yangxin decoction treatment group,captopril treatment group and yangxin decoction prevention groupwere lower(P<0.05).Conclusion:1.Yangxin decoction can improve the indexes of hemodynamics and the cardiac function of the rats with chronic heart failure.2.Yangxin decoction have the effect of myocardial preservation through reducing the myocardial pathological damage,and improving the myocardial ultrastructure of the organization of the rats with chronic heart failure.3.Yangxin decoction can inhibit the myocardial remodeling through reducing the concentration of Ca2+ and CaN,down-regulating NFAT3 mRNA、GATA4 mRNA expression,decreasing NFAT3、DATA4 protein expression and inhibiting the Ca2+-CaN-NFAT3 signal pathway.
