EFFECTS OF ANTIEPILEPTIC DRUGS ON BEHAVIOR AND SYNAPTIC PLASTICITY IN SUBTHRESHOLD CONVULSANT DISCHARGES MODEL RATSObjective:To investigate the effects of antiepileptic drugs on behavior and synaptic plasticity in subthreshold convulsant discharges(SCD)model rats.Methods : Subthreshold convulsant discharges(SCD)model was established by electrical stimulation of the hippocampus,the rats were subjected to sodium valproate and levetiracetam gavage.Behavior of rats was observed by Morris water maze.Synaptic plasticity was observed by Long-term potentiation and neural cell adhesion molecule(NCAM),postsynaptic density-95(PSD-95),growth associated protein-43(GAP-43)expression levels.Results:The spatial learning ability and spatial memory ability of SCD model rats were impaired,the slope of the hippocampal field excitatory postsynaptic potential(f EPSP)in SCD model rats was decreased.The level of PSD-95 in hippocampus of SCD model rats was decreased,and the levels of NCAM and GAP-43 were increased.After levetiracetam(LEV)intragastric administration,the spatial learning ability and spatial memory ability of SCD model rats is better than sodium valproate group.Levetiracetam can increase the slope of hippocampal f EPSP in SCD rats,levetiracetam can reduce the level of hippocampal NCAM and GAP-43 in SCD model rats.Conclusions: Subthreshold convulsant discharge can impair cognitive functions of rats.LEV can improve cognitive function in SCD model rats.NCAM,PSD-95 and GAP-43 may be involved in the improvement of cognitive function.EFFECTS OF LEV ON PKC ACTIVITY AND CAMK?? IN HIPPOCAMPUS OF SCD MODEL RATSObjective:To investigate the effects of LEV on PKC activity and Ca MK?? in hippocampus of SCD model rats.Methods : Promega reagent kit was used to test the level of phosphorylated PKC in rat hippocampus.Using Western-Blot and RT-q PCR to test the level of Ca MK?? in rat hippocampus.Results:The level of phosphorylated PKC and Ca MK?? expression in hippocampus of SCD model rats was lower than that in control group.After intragastric administration of levetiracetam in SCD model rat,the level of phosphorylated PKC and Ca MK?? expression is higher than SCD model group.Conclusions: PKC-GAP43-Ca MK pathways may be involved in the cognitive function of LEV to improve the SCD model rats.STUDY ON THE TREATMENT OF ANTI EPILEPTIC DRUGS IN PATIENTS WITH SUBCLINICAL EPILEPTIFORM DISCHARGEObjective:To investigate the clinical efficacy of treating subclinical epileptiform discharge patient with antiepileptic drugs.Methods:(1)A retrospective analysis of sodium valproate combined with atomoxetine treatment of attention deficit hyperactivity disorder(ADHD)comorbid subclinical epileptiform discharge(SED).(2)A retrospective analysis of levetiracetam treatment of autism spectrum disorder(ASD)comorbid subclinical epileptiform discharge.Results:(1)The efficacy of treating ADHD comorbid SED with atomoxetine plus sodium valproate is similar to that of monotherapy with atomoxetine.(2)The efficacy of treating ASD comorbid SED with levetiracetam plus special education is similar to that of monotherapy with special education.Conclusions: Antiepileptic drug treatment in patients with subclinical epileptiform discharge did not find a significant effect.
