Background Liver malignancies like hepatocellular carcinoma and intrhepatic cholangiocarcinoma has become a worldwide health problem for their high morbidity,bad biological behavior and resistance to radiotherapy and chemotherapy.Partial hepatectomy is the main effective method to liver malignancies and other liver diseases.Extend partial hepatectomy is peformed to get better prognosis when huge liver cancer or tumor invading main liver vessels occured.Sometimes,functional liver volume deficiency is accompanied by extend partial hepatectomy and thus may result in post hepatectomy liver failure.Underlying diseases like alcolholic liver disease and cirrhosis generate higher risk to extend partial hepatectomy together with higher morbidity of liver malignancies.Recently,as the research progress of stem cell and liver regeneration,it is proved that CD133 stem cell can join in the process of liver regeneration under special situation.In this research,85% partial hepatectomy was performed on Lewis rats to find the effect of CD133 stem cell during liver regeneration and the impact of alcoholic injury on post hepatectomy liver regeneration.Objective To find the effect of CD133 stem cell on post 85% partial hepatectomy liver regeneration on Lewis rats.To indentify the effect of pharmacal mobilization of stem cell on Lewis rats post 85% partial hepatectomy.To build a rat animal model mimicking human alcohol abuse pattern.To investigate the effect of alcoholic injury on post extend hepatectomy liver regeneration.Methods1.Observe the process of liver regeneration and the effect of CD133 stem cell on male Lewis rats received 85% partial hepatectomy and AMD3100 combined low dose tacroliums stem cell mobilization treatment.2.Build up chronic plus binge alcohol intake animal model on male Lewis rats,then perform 85% partial hepatectomy and observe the process of liver regenerationResults 1.85% partial hepatectomy generated severe liver injury on Lewis rats,more CD133 stem cells were found in the liver during liver regeneration.Stem cell mobilization further increased CD133 stem cell number together with proliferating cell number.2.Chronic plus binge animal model generated obvious liver injury and steatosis.85% partial hepatectomy on alcoholic liver injury animal generated extreme liver injury.Proliferating cell number at early post operation stage is obviously lower than controlConclusion 1.After 85% partial hepatectomy,SDF-1/CXCR-4 signal pathway contributes to stem cell mobilization,CD133 stem cell joins in the process of liver regeneration.2.AMD3100 combined low dose tacrolimus treatment enhances stem cell mobilization effect,improves liver regeneration after 85% partial hepatectomy in a safe and effective method,which is of great clinical potential.3.Alcoholic liver injury will dramatically increase post hepatectomy liver injury,inhibit early stage liver regeneration and postpone the regeneration peak.
