Experimental Study On The Effect Of Warming Kidney And Reinforcing Deficiency Prescription On Hypothalamus-pituitary-Thyroid/gonadal Axis Function In Hypothyroid Rats

Objective: Experimental study on the effect of warming kidney and reinforcing deficiency prescription on hypothalamus pituitary thyroid gland axis function in SD rats.Methods: 60 male SD rats were selected and fed for a period of 1 weeks.The rats were randomly divided into the normal group and the experimental group.The rats in the experimental group were treated with PTU solution(distilled water with a concentration of 0.05%),and the rats were continuously made for a period of 4weeks.The rats were randomly divided into kidney tonic low dose group(referred to as low temperature group),warming kidney and reinforcing deficiency prescription group deficiency(Wen Zhongzu),kidney tonic high dose group(Wen Gaozu),western medicine group,model group,10 rats in each group.Each group was given the corresponding medicine,the model group and the normal group was given distilled water for 28 days.The general situation of rats in each group was observed during the experiment.After the intervention,were detected in hypothyroid rats,serum thyroid hormone levels,observe the histopathological changes of the pituitary gland,thyroid gland and testis;expression using RT-PCR technique for determination of telomerase catalytic subunit,Bcl-2,testis Fas gene;by immunohistochemical determination of pituitary,thyroid,testicular tissue and thyroid receptor protein expression method.Results: In this study,the rat model of hypothyroidism was successfully established,and the pathological results showed that the pathological changes were in accordance with hypothyroidism.warming kidney and reinforcing deficiency prescription can improve the symptoms of hypothyroidism rats,which is more obvious than that of Western medicine group.warming kidney and reinforcing deficiency prescription can improve the rats eating,water intake,body weight,Anal temperature,etc.,which is more obvious than that of Western medicine group.The method of warming kidney and reinforcing deficiency prescription can improve thelevel of thyroid hormone in hypothyroidism rats,which is more obvious than that in western medicine group.The prescription of warming kidney and reinforcing deficiency prescription can improve the damage of pituitary gland,thyroid gland and testicle tissue of rats with hypothyroidism,which is more obvious than that of Western medicine group.The kidney tonic side of thyroid hormone receptor expression in testis of rats can increase hypothyroidism,is more obvious than western medicine group,the middle dose group was the best.The kidney tonic can promote Bcl2 and increase the expression of TERT,inhibit the expression of Fas was more obvious than western medicine group,the middle dose group was the best.Conclusion: warming kidney and reinforcing deficiency prescription can improve the symptoms of hypothyroidism rats and thyroid hormone levels,improve thyroid morphology,promote the recovery of thyroid function,but also through the thyroid hormone receptor expression in testicular tissue,promote Bcl2 and increase the expression of TERT and inhibition of Fas expression,thereby inhibiting the apoptosis of testicular cells,promote the recovery of gonadal function.
