Background:The incidence of gastrointestinal stromal tumor is increasing year by year.Early detection is extremely difficult and the rate of misdiagnosis is high.The recurrence rate and drug resistance rate of target treatment increased gradually.The treatment is faced with a bottleneck.Ghrelin is a 28 amino acid acylated peptide,mainly produced by the stomach fundus,and can promote the proliferation of malignant tumors.Ghrelin may provide new ideas for the diagnosis and treatment of GISTs.Objective:1)whether Ghrelin can be used as an indicator of GISTs to assist in diagnosis and prognosis.2)The preliminary mechanism of Ghrelin participates in GISTs growth regulation;3)whether Ghrelin can be used as a reference index for nutritional risk assessment of gastrointestinal cancer during perioperative period.Method:1)collect the serum of normal persons,GISTs and patients with digestive tract malignant tumor and measure serum ghrelin.GISTs tumor and paracancerous normal mucosa tissues were collected and ghrelin and its receptor contents were measured.We associate ghrelin with clinicopathological data to analyze the correlation between ghrelin and GISTs and its role in nutritional risk assessment.2)In vitro cell immunohistochemistry,cell proliferation assay and Western blotting were used to explore the mechanism of ghrelin regulating GISTs growth.Result:1)serum Ghrelin can be used as a reference index for the existence of GISTs,but it has no specificity.It can't judge the prognosis of the patient.2)tissue Ghrelin can be used as a reference index for GISTs malignancy and recurrence risk,3)GISTs secrete Ghrelin through autocrine and paracrine.4)Ghrelin and its receptor antagonists can induce ghrelin receptor expression.5)Ghrelin participates in the regulation of GISTs growth through multiple pathways.6)Ghrelin can be used as a useful reference index for judging nutritional risk.Conclusion:Ghrelin participates in the regulation of GISTs growth and has a certain reference value for the malignant degree of GISTs,the risk of recurrence and the perioperative nutritional risk assessment of the patients with digestive tract malignant tumor.
