Study On The Treatment Mechanism Of Intestinal Flora Disturbance And Its Metabolite Lactic Acid Protecting Intestinal Mucosa In Children With Allergic Purpura With DaHuang Mudan Decoction

ObjectiveAnaphylactoid purpura(AP),also known as henoch-schonlein purpura(HSP),is the most common small vessel inflammation as the main lesion of allergic disease in children.Because the pathogenesis is not clear at present,the therapeutic effect and mechanism of this disease are affected.Studies have shown that children with allergic purpura have different degrees of intestinal flora disorder,which may be related to its pathogenesis and mechanism.This study based on the long-term clinical effect of TCM treatment of allergic purpura observation and experience,explore classical compound rhubarb soup of cortex moutan disorders in children with allergic purpura occurred intestinal bacterial species distribution of specific impact,how to adjust the HSP strains of intestinal disorders,reshape the intestinal bacterial species distribution,and how to affect intestinal mucosal inflammatory immune signal transduction,so as to improve the function of intestinal mucosa,relieve the clinical symptoms of HSP.The possible mechanism of treating allergic purpura with rhubarb mudan skin decoction was studied in depth.The adjustment of intestinal flora and the protection of intestinal mucosa may be a new idea for clinical treatment of HSP,providing a better diagnosis and treatment basis for the pathogenesis,prognosis and prognosis of HSP diseases and providing an effective new treatment scheme.MethodsUsing Elisa,high-throughput sequencing technologies,collect health group,HSP mild,HSP severe three groups of children before and after treatment in the rhubarb cortex moutan soup excrement and urine and blood,combined with clinical symptoms and laboratory indications of changes associated with disease,the research on the clinical symptoms of HSP rhubarb soup of cortex moutan,the improvement of the intestinal mucosa damage degree and function disorder,adjust the structure of flora distribution found HSP intestinal flora disturbance on the strain distribution rule and rhubarb cortex moutan soup adjustment,found that the drug sensitive probiotics;Then,in vitro culture of lactobacilli,which is sensitive to drugs,was studied to explore the rule of action of rhubarb peony bark decoction on strains.The metabolic products of probiotics after drug intervention were applied to isolated intestinal epithelial cells,and the mechanism of protecting intestinal epithelial cells was explored by using immunofluorescence technology.Results1.Intestinal flora disorders did occur in children with allergic purpura,mainly gram-negative bacteria;Rhubarb peony bark decoction has the function of redistributing intestinal flora,in which lactobacillus is drug sensitive probiotics.2.Da Huang Mu dan decoction can promote the proliferation of lactobacillus and release more acid metabolites lactic acid.3.Lactate can improve the balance of target protein in lps-induced inflammatory immune signal nf-kb pathway of intestinal epithelial cells,and has the therapeutic effect of promoting cytoskeletal protein and tight junctions.ConclusionDa Huang Mu dan decoction can regulate the intestinal flora structure of children with HSP and then remodeling,which can promote the metabolites of sensitive probiotics lactobacillus,and its metabolite lactate can improve the inflammatory immune function of intestinal mucosa and protect the integrity of intestinal mucosa.
