ObjectiveTo summarize the clinical experience of Professor Yang Liu in the treatment of melanin metabolic disorders by using the method of regulating qi and blood;to study the effect of melanin metabolism by Chinese herbal medicine under the guidance of harmonized qi and blood method on the experimental model of zebrafish;study the effect of Chinese medicine on the melanin metabolism of human melanin HEM-d cells;study the effective components and targets of tribulus terrestris on melanin metabolism regulation were studied from a holistic perspective.Method1.Through the follow-up study,summarize the clinical experience of Professor Yang Liu using the method of regulating qi and blood to treat melanoma metabolic disorders,especially to summarize Professor Yang Liu’s clinical experience in treating chloasma.2.The traditional Chinese medicine under the guidance of qi and blood method was applied to the zebrafish juveniles to calculate the melanin area of the specific part of the head.The melanin content was detected by NaOH method,and the tyrosinase activity was detected by dopa rate oxidation method.Western Blot method was used to detect the melanin content.Protein expression of c-kit,MITF,TYR,TYRP-1,and mRNA expression of MITF-a,MITF-b,c-kit,TYR,TYRP-1,and MITF-a were detected by qPCR.3.20 common Color Chinese medicines were applied to human melanin HEM-d cells.The cell proliferation activity was detected by MTT assay,the melanin content was detected by NaOH method,and the tyrosinase activity was detected by dopa rate oxidation method.4.Using the pharmacology of traditional Chinese medicine to analyze the chemical constituents and target of melanin in Chinese herbal medicine,then establish a component-target-disease model,and explore the mechanism of hedgehog regulation of melanin metabolism.Results1.Clinical experience from Professor Yang Liu’s in the treatment of melanin metabolic disorders:Professor Yang Liu treats melanin metabolic disorders with the method of regulating qi and blood,specially through these three specifics of"dispelling the wind,promoting blood circulation,replenishing liver and kidney".2.In the in vivo experiment of zebrafish model,the Chinese herbal medicine of tribulus terrestris under the guidance of the hedgehog and the qi and blood method inhibited the melanin content and tyrosinase activity of the zebrafish juveniles at a certain concentration,and it has not been observed that the tribulus terrestris has a two-way regulation.The tribulus terrestris combination group promoted the mRNA expression of MITF,c-KIT,TYRP-1 and MITF-a(P<0.05).3.The color Chinese medicine of each group has different effects on the regulation of melanin content and tyrosinase.Nine of the traditional Chinese medicine of deep color generally has an activation effect,and eight of the traditional Chinese medicine of light color generally has an inhibitory effect.4.Obtain the active constituents,target targets,related action pathways and biological processes of tribulus terrestris on melanin regulating metabolism.Conclusion1.Treating melanin metabolic disorders with the method of regulating the qi and blood has clinical application significance.2.The compatibility of traditional Chinese medicine under the guidance of the qi and blood method has a more obvious effect on the metabolism of melanin,and also has a tendency of bidirectional regulation of melanin.3.Deep color Chinese medicine generally has an activation effect,and light color Chinese medicine generally has an inhibitory effect.4.The regulation of melanin by hedgehogs is related to multiple pathways and biological processes,but the specific mechanism of bidirectional regulation has not been explored.
