Objective:This study applied single acupoint and acupoints combination acupressure in the treatment of functional dyspepsia model rats,combined the technology of metabonomics and UPE,by comparing the observed before and after treatment of functional dyspepsia rats metabolites content and specific parts UPE intensity change,scientific evidence the validity of the Tuina acupressure therapy in the treatment of functional dyspepsia,preliminary discussion on application of different acupoints differences in treatment of functional dyspepsia effect mechanism,providing scientific basis for the Tuina clinical treatment and choice of acupoints.Methods:This study is an animal experimental study to discuss the modeling quality of functional dyspepsia rats from gastrointestinal dynamics and histomorphology.The effectiveness of acupressure therapy and the difference between single acupoint and acupoints combination therapy were discussed by means of two objective indexes: metabolomics and ultra-weak luminescence intensity.Sixty SPF healthy Wister rats were randomly divided into five groups,namely the blank group,the model group,the acupoint matching group,the zhongwan group and the weishu group,with 12 in each group.In addition to the blank group of 12 rats,the other 48 rats were modeled.The rat model of functional dyspepsia was prepared by the method of compound etiology modeling(including clip-tail stimulation and irregular diet)for 14 days.After the modeling,10 rats were randomly selected from each group and their stomachs were dissected to observe the stomach and duodenum of the rats.HE staining sections were made to confirm that there were no organic lesions in the rats,and the dynamic function of the gastrointestinal tract was detected to verify the modeling quality of the rats with functional dyspepsia.Starting from the 3rd week of modeling,the model group received fixed grasping stimulation every day.In the acupoint group,the massage was treated with zhongwan and weishu points;in the zhongwan group,the massage was treated with zhongwan points;in the weishu group,the massage was treated with weishu points.During the experiment,the weight and food intake of the rats were measured,and the changes of the rats’ posture and behavior were observed.After treatment,rat abdominal aortic serum was extracted and analyzed based on ultra-high performance liquid chromatography-q-tofu MS.Three rats were randomly selected from each group for ultra-weak luminescence detection to observe the effects of different acupoints combined with massage and acupoint therapy on serum metabolites of functional dyspepsia rats.Using positive and negative ion metabolites and their possible associated pathways as the mechanism of action,the differences of the mechanism of functional dyspepsia by different acupoints were analyzed.Results:1.Modeling results of functional dyspepsia rats:The weight records of each group were analyzed and no significant difference was found in each group before modeling(P > 0.05).After 2 weeks of modeling,the model group,matched acupoint group,zhongwan group and weishu group were compared with the blank group in weight loss and showed significant differences(P < 0.01).After the treatment,the model group lost weight and showed differences with that of the matching point group,the zhongwan group,the weishu group and the blank group(P < 0.05).There was no significant difference in body weight between the acupoint matching group and the zhongwan group and the weishu group(P > 0.05).Gastrointestinal dynamic test showed that after 2 weeks of modeling,the gastric residual rate of the model group was significantly increased and the intestinal propulsion rate significantly decreased compared with the blank group(P < 0.01),as well as the matching point group,the zhongwan group and the weishu group.HE staining and sections of rat stomach tissue showed that there were no organic gastric lesions in the model group,the matching point group,the zhongwan group and the weishu group.2 metabolomics results:The quality control experiment shows that the instrument analysis system has good stability and the test data is stable and reliable.The difference of metabolic spectrum obtained in the experiment can reflect the biological difference between samples.There are differences between the groups of serum metabolites are both positive and negative ions,by Fisher’s exact test method for each group of differentially expressed metabolites KEGG pathway enrichment analysis,the results show that the model group compared with blank group,Protein digestion and absorption,Central carbon metabolism in cancer,Aminoacyl tRNA-biosynthesis,Mineral absorption,Significant changes have taken place in important pathways such as ABC transporters and Arginine biosynthesis.Model group compared with group with acupuncture point,Protein digestion and absorption,Central carbon metabolism in cancer,Aminoacyl tRNA-biosynthesis,Mineral absorption,ABC transporters,and other important pathways changed dramatically;Chung wan and wei shu group compared with group with acupuncture point,Protein digestion and absorption,Central carbon metabolism in cancer,Aminoacyl tRNA-biosynthesis pathways such as changed.3.Ultra-weak luminescence intensity test results: the ultra-weak luminescence intensity(PMT)test results showed that after treatment,the light-emitting enhancement of the acupoint matching group,the zhongwan group,the weishu group and the blank group were significantly different from the model group(P < 0.01).Compared with the zhongwan group and the weishu group,the light emission in the acupoint matching group was enhanced and there was a difference(P < 0.05).Conclusion:1.Through the comparison of functional dyspepsia rats before and after Tuina acupressure treatment,the therapeutic effect of Tuina acupoint therapy in promoting gastrointestinal motility is accurate,which proves the effectiveness of massage acupoint therapy in the treatment of functional dyspepsia.2 through metabonomics studies have demonstrated that a series of positive and negative ions metabolites deviating from the norm is the common characteristic of functional dyspepsia rats serum metabolism,single cavity and compatibility with acupuncture acupoints can affect a variety of important pathways through purpose of treatment,single cavity and compatibility with acupuncture acupoints in treatment of functional dyspepsia rats influence related pathways is different,its effect mechanism have certain differences.3 by ultra weak luminescence intensity test proves that the single acupoint and acupoints combination with Tuina acupressure therapy can significantly enhance the functional dyspepsia rats stomach ultra weak luminescence intensity(PMT),metabonomics analysis shows that massage therapy mechanisms may be related to protein digestion and absorption,membrane transport,various amino acid synthesis and energy change.Different compatibility with acupuncture acupoints massage therapy on functional dyspepsia rats and the effect of the ultra weak luminescence intensity has certain differences,with acupuncture point group of ultra weak luminescence intensity is stronger,illustrate acupoints compatibility treatment effect mechanism may be associated with more energy change,for the future similar ultra weak luminescence study provides reference and reference.4.Through this study,on the one hand,it is proved that the compatibility of acupoints has an important guiding role in clinical massage,and zhongwan and weishu acupoints have a good therapeutic effect in the treatment of functional dyspepsia.On the other hand,it has been proved that the effect mechanism of treating functional dyspepsia by the compatibility of acupoints and single acupoint is closely related to various important pathways,and the effect mechanism of applying different acupoints is different to some extent.From the perspective of clinical and biological mechanism,it provides scientific basis for the clinical treatment and acupoint selection of massage.
