This article was divided into three parts.The first part reviewed the theoretical source of the"Yin-Yang Rizhong adjustment method",and summarized the chapters of"Huang Di Nei Jing"’s thinking about the"Correspondence between Heaven and human".The second part was the first experiment.It explored the effects of time-delayed full-spectrum halogen lamps on the development of young rabbit eyes.The serous dopamine,retinal dopamine and circadian clock gene Cry expression were used as indicators to investigate the influencing mechanism and intervention mechanism of traditional Chinese medicine.The third part was the second experiment.It explored the effect of time-delayed irradiation of full-spectrum halogen lamps on the expression of opsin and the ultrastructure of retina,and discussed the intervention of traditional Chinese medicine.The following describes the contents of the two parts of the experiment.Experiment 1Objective:To investigate the effects of time-delayed full-spectrum halogen lamps on the development of eyeballs in young rabbits and the intervention mechanism of traditional Chinese medicine.Methods:54 New Zealand white rabbits aged 4 weeks were randomly selected and divided into 3 groups:natural light group,full-spectrum group and full-spectrum Chinese medicine group.The natural light group was placed about 1m by the southern window.The full-spectrum group and the full-spectrum Chinese medicine group were placed about 60cm in front of the quartz halogen lamp frame,and there was no other interference light source during the experiment.The artificial light source group was set at the timer 6:00 starts lighting and 23:59 turns off lighting every day.The young rabbits of the Chinese medicine group were given the "Ri Zhong Yang formula" daily at 10:00,and were given the "Ri Zhong Yin formula " daily at 22:00.The experiment was conducted for a total of 12 weeks.Eye axis leng ths were measured at the 4th,8th,and 12th week of the experiment,and the retinas of the young rabbits were collected at the same time.The relative contents of Cryl and Cry2 proteins in each sample were detected by Western blot,and Cry1 mRNA and Cry2 mRNA expression were detected by RT-PCR.At 4 and 12 weeks of the experiment using enzyme-linked immunosorbent assay to measure the dopamine content of each serum sample.The content of dopamine in each retina sample was analyzed by high performance liquid chromatography.RESULTS:Eye axis:At the 4th and 8th week of the experiment,the axial length of young rabbits in the full-spectrum group was longer than that in the natural light group and the Chinese traditional medicine group(P<0.05).At week 12 of the experiment,The full spectrum group was longer than the other two groups,but the difference was not statistically significant(P>0.05).Serum dopamine:At the 4th and 12th week of the experiment,the natural light group and the full spectrum Chinese medicine group were higher than the full spectrum group,but the difference was not statistically significant(P>0.05).Retinal dopamine:At the 4th and 12th week of the experiment,the content of the full-spectrum group was lower than that of the natural light group and the full-spectrum Chinese medicine group,and there was a statistically significant difference from the natural light group(P<0.000),and it was no significantly different from the full-spectrum Chinese medicine group(P>0.05).Relative quantification of Cry1 protein:At the 4th,8th,and 12th week of experiment,the expression of Cry1 protein in the retina of the rabbits in the full-spectrum group was lower than that in the natural light group and the traditional Chinese medicine group,all the differences were statistically significant(P<0.05).Relative quantification of Cry2 protein:At the 4th and 12th week of the experiment,the expression of Cry2 protein in the retina of the full-spectrum group was lower than that of the natural light group and the full-spectrum Chinese medicine group(P<0.000);at the 8th week of the experiment,the Cry2 protein expression in the retina of the full spectrum group was higher than that in the natural light group and the full-spectrum Chinese medicine group,the difference was statistically significant(P<0.05).Cry1 mRNA expression:At the 4th,8th,and 12th week of the experiment,the expression of Cry1 mRNA in the retina of the full-spectrum group was lower than that of the natural light group and the full-spectrum Chinese medicine group,and the difference was extremely significant(P<0.000).Cry2 mRNA expression:At the 4th,8th,and 12th week of the experiment,the expression of Cry2 mRNA in the retina of the full-spectrum group was lower than that of the natural light group and the full-spectrum Chinese medicine group,and the differences were statistically significant(P<0.05).Conclusions:1.Delayed irradiation of the full-spectrum halogen lamp may cause the axial axis of young rabbits to accelerate to myopic development to a certain extent,which may be related to the decrease of retinal dopamine levels.2.The growth of young rabbits in the delayed irradiation environment can cause the disorder of retinal clock transcriptional regulator Cry expression.3.Chinese medicine can inhibit the acceleration of myopia caused by delayed irradiation of full-spectrum halogen lamps by promoting the release of dopamine.4."Ri zhong Yin Yang formula" has a certain adjustment effect on Cry disorders.Experiment 2Objective:To explore the difference of opsin expression and the ultrastructural changes of the retina in rabbits exposed to full-spectrum halogen lamps with time-delayed irradiation,and to discuss the effects of intervention of traditional Chinese medicines.Methods:54 New Zealand white rabbits aged 4 weeks were randomly selected and divided into natural group,full-spectrum group and full-spectrum Chinese medicine group.The experimental light environment setting and traditional Chinese medicine administration method were the same as in the first experiment.The experiment was conducted for a total of 12 weeks.The retinas of each group of rabbits were collected at the 4th and 12th week of the experiment,and the L-opsin,M-opsin and S-opsin were detected by Western blotting.The mRNA expression of the three kinds of opsins was detected by RT-PCR.At 4 weeks,8 weeks and 12 weeks of experiment,retinas of the rabbits were subjected to electron microscopy.Results:Expression of opsin and mRNA:At the 4th and 12th weeks of the experiment,the expression of L-opsin and M-opsin was lower in the whole-spectrum group than in the natural light group and the full-spectrum Chinese medicine group,and the difference was statistically significant(P<0.05).In the 4th week,the S-opsin expression in the full spectrum group was higher than that in the natural light group,which was lower than that in the full spectrum Chinese medicine group.The difference was statistically significant(P<0.05).At the 4th and 12th week of experiment,the mRNA expression of the three opsins in the full-spectrum group was lower than that in the other two groups,except for the 12th week when compared with the full-spectrum Chinese medicine group,there was no statistical significance(P<0.05).Electron microscopic results:At the 4th week of the experiment,no obvious pathological changes were observed in the three groups.At the 8th week of the experiment,the microvilli on the photoreceptor surface of the retinal pigment epithelial cells in the full-spectrum group decreased,and larger lipid droplets were seen in the cytoplasm.The number of mitochondria in the inner segment of the ellipse was reduced,and occasionally large vacuoles were occasionally observed between the photoreceptor cells.The photoreceptors of the full-spectrum Chinese medicine group had normal outer membrane structure,occasionally vacuoles in the membranous disc,and some mitochondria in the inner segment oval were loosely arranged.cut back.At the 12th week of the experiment,the structure of the retina layers in the full-spectrum group was unclear,and only the residual photoreceptor outer ganglion disk and the outermost scleral structure were seen.The outer peritoneal disk of the full-spectrum Chinese medicine group had a loose structure and the mitochondria in the inner ellipsoidal body.amount reduced.Conclusion:1.The early L-opsin and M-opsin expression of rabbits growing under the full-spectrum halogen lamp was lower than that of young rabbits grown under natural light.The expression of S-opsin was higher in the former than in the latter,under the full-spectrum halogen lamp.The growth of young rabbits had significantly lower levels of three opsins than young rabbits grown under natural light.2.In the short term,the full-spectrum halogen lamp had no obvious damage to the ultrastructure of the retina,but long-term irradiation showed damage to the structure of the retina.3."Ri Zhong Yin Yang Formula " has a certain up-regulated expression of three kinds of opsins in young rabbits grown under the full-spectrum halogen environment,and it has a certain protective effect on long-term prolonged exposure to retinal damage.
