Objective:To indentify the most significant different metabolites between higher and lower cardiorespiratory fitness?CRF?,pre-exercise and post-exercise in prediabetes?PDM?.To provide theoretical and practical basis for the mechanism of exercise intervention in PDM.Part 1Methods:80 PDM subjects were selected.Body composition measured by dual-energy x-ray absorptiometry.CRF was measured directly with a graded exercise test.Plasma metabolites were analyzed using ultra high performance liquid mass spectrometry?UPLC-MS?.PCA and OPLS-DA were used to analysis different metabolites.Results:Compared with lower CRF group,in higher CRF group,HOMA-IR,HOMA-?is lower,LDL-C,BMI and Fat%is lower,low intensity activity?LPA?is more.The metabolic characteristics were significantly different higher and lower CRF,there were 7 metabolites,including PC?20:1/14:1?,PC?18:3/16:0?,LysoPC?16:0?,Valine,isocitric acid,Octyl carnitine and Linoleyl carnitine.Part 2Methods:60 PDM subjects were selected,and received exercise or health education for 12 weeks.Exercise training:3 times/week,50 min per session at 46%-64%VO2max.Results:After exercise training,61.11%blood sugar turn normal,total energy expenditure?TEE?,physical activity energy expenditure?PAEE?,medium to large intensity activity?MVPA?,LPA were increased,2hPG,triacylglycerol were decreased.The metabolic characteristics were significantly different before and after exercise,there were 15 metabolites,including LysoPC,PC,phytosphingosine,sphingosine,hexadecylsphingosine,valine,leucine.Conclusion:?1?PC,LysoPC,valine,isocitrate and acylcarnitine may be potential biological markers of CRF.?2?Exercise intervention influenced the metabolism of phospholipid and branched chain amino acids,it may be one of the mechanisms to improve the metabolism of glucose and lipid in PDM group.?3?The effect of exercise intervention on plasma metabolites in PDM subjects with different CRF still needs further improvement and demonstration.
