TypeⅠinterferons containing multigenes , plays a significantly important role in terms of the antiviral , anti-microbial , anti-cell proliferation functions.Thymosin alpha1 (Talpha1) is a 28-amino acid peptide,a well-known immune system enhancer for the treatment of various diseases.Evidence has been provided that combined treatments with interferon (IFN) and thymosin alpha 1 are highly effective.At present,there are mainly combination with interferon (IFN) and thymosin alpha 1.But there were few reports about expression interferon (IFN) and thymosin alpha 1 as a integrated protein.In this study,We design a flexible oligopeptide gene linker contain 5 amino acids,IFNα1-THYα1 was conjunction together by this linker.we obtain the fusion gene by PCR.we expression interferon (IFN) and thymosin alpha 1 recombinant gene,and get the fusional protein.The expression plasmid pGEX4T-2 Vector was used as the vector to construct the expression plasmids pGEX4T-2-IFNα1-THYα1 by technology of gene engineering.The recombinant plasmids were transfected into BL21(DE3)cells,induced by IPTG at 37℃,transcript and expression.And there was a 43KDa protein in polyacrylamide gel electrophoresis.The results showed that the recombinant plasmids could express the interest proteins.We obtain the pure protein through inclusion by Walterson GST-Resin Kit. We have detect the active of the pure protein,The results showed that this fusional protein has anti-virus activity of IFNα1 and it also has the activity of thymosinα1.
