Objective: Implantation is a complex developmental process. It contains a series of synchronous and concordant variations which happened between embryo and uterus. Implantation dates from the recognization, localization, adhesion between nomadic blastocyst and uterine epithelium, soon after tropHEC-1A-1Atoderm cell penetrate the surface of endometria and embed into the groundsubstance.Lewis Y(LeY)antigen is a double-fucosylation oligosaccharide. LeY antigen exists in embryo cell, endothelial cell and neutrophilic granulocyte. LeY antigen was catalyzed by FUT4, which is a key enzyme in LeY synthesis.Study the expression of FUT4 and LeY antigen, compare and analyze FUT4 expression on endometrial cell.Methods: The experiment choose high-receptive (RL95-2) and low-receptive (HEC-1A) epithelial endometrial cells lines. Detect the expression of FUT4 on endometrial cell by RT-PCR and Western blot; in cell transfection, FUT4 siRNA was transfected to RL95-2 cells and FUT4 over-expression plasmid was transfected to HEC-1A cells, in order to check out the expression of FUT4 and LeY on endometrial through immune- ohistochemisty and cell adhesion experiment; meanwhile in cell adhesion experiment, LeY antibody was added in two different condition endometrium cell to observe the adhesion rates between two endometrial cell(RL95-2 cell and HEC-1A cell) and embryo cell(JAR cell).Results: FUT4 and LeY in RL95-2 cells were higher than HEC-1A cells by Western blot and RT-PCR; cell adhesion experiment indicated that the adhesion ratio between embryonic cell and RL95-2 was higher than with HEC-1A; Through immunohistochemisty experiment, we found that exp- ression of FUT4 and LeY decreased if FUT4 siRNA was transfected to RL95-2 cells; LeY and FUT4 increased when FUT4 over-expression plasmid was transfected to HEC-1A cells. Blocked with LeY antibody, the adhesion ratio of JAR cells with RL95-2 cells or HEC-1A cells were all significant decreased.Conclusion: The result indicate that LeY antigen is higher expressed in high-receptive cells, the level of FUT4 and LeY correlate with the receptivity of endometrium cells. It may supply the way of interfering embryo implantation by carbohydrate mediated approaches.
