Digital holographic microscopy combines the digital holography and holographic microscopy, and realizes the process of holographic microscopy with CCD to replace the traditional to the holographic plate. Digital holographic microscopy comparing to the traditional microscopy has many merits, such as recording and reconstruction three-dimensional information of objects, high resolution, the smaller impacting on samples, simple equipments, and so on. So it is widely used in biological cells, micro-particle imaging and tracking, the growth of polymer particle detection, detection of micro-circuits and other fields.The paper basing on the principle of optical holography, introduces the principle of recording and reconstruction of the digital holography. In this paper, the conditions of the digital holographic experimental are analyzed and discussed. The paper studies the basic theory of the transmission Fourier-transformed digital holography, designs the experiment, and reconstructs the image of the objects using the Fourier transform method. We discuss the digital two types of zoom mode of the holographic microscopy, and focus on the pre-amplification Fresnel off-axis holographic microscopy. We also design the optical path of the pre-amplification Fresnel off-axis holography, and reconstruct the image of onion cells and the stem of lily cells. At last we measure the size of the paramecium by calibrating the resolution target.
