FU ZHENG HUA YU, a traditional Chinese medicine (TCM) formula for hepatic fibrosis disease, is composed of Salvia miltiorrhiza Bge, Cordyceps Sinensis, Semen Persicae, Gynostemma Pentaphyllum. This paper mainly studied the process of both extraction and purification of active constituents in Semen persicae and Salvia miltiorrhiza Bge.The chemical basis of Semen Persicae was studied. The major peaks of HPLC were identified by LC-MS technique, and amgydalin components were defined as the effective components. Conditions of extraction and purification of amgydalin components from Semen persicae were investigated with the single-factor method. The optimized extraction process was extracted for 90 min using 10 times of 50% ethanol solvent. The separation procedure was perform on a porous adsorbent of HZ 818 resin with sample loading rate of 1 BV/h, and then the effective components were eluted with 50% ethanol. The yield of the amgydalin components was about 0.81%, and the content of the amgydalin components was about 63%. The process of extraction and separation was enlarged and the results showed that this method was stable and reliable.Conditions of extraction and purification process were selected with the yield of the phenolic acids components. Purifition of the phenolic acids components in the extract-liquid with macroporous resin was studied and the technological parameters were selected. The optimized extraction process was extracted twice for 60 min, using 10 times of water once, mergered the extraction liquid. The separation procedure was perform on a porous adsorbent of HZ 816 resin with sample loading rate of 1/2 BV/h, and then the effective components were eluted with 40% ethanol. The yield of the phenolic acids components was about 8.3%, and the content of the phenolic acids components was about 25%. The process of extraction and separation was enlarged and the results showed that this method was stable and reliable.
