This paper has researched on the metabolic mechanism and the method of purification of an antibiotic substance produced by Rhizopus chinesis 12th.Applying of stimulating method and static cell method ,we observed that acetate, histidine and arginine have promotive effects on the production of antibiotic substance when they were added to culture. By added protein biosynthesis inhibitor-cycloheximide to the culture medium, we discovered that the biosynthesis of the antibiotic substance has relations with synthesis of the mycelial protein. This property is different from that of the ordinary peptide antibiotic. The probably metabolic pathways was conjectured on the basis of experiment conclution.The crude extrate of antibiotic substance was stable in prepare experiment, so we can isolate this substance on commomly condition instead of on special protective measures.The proper method of purifying the antibiotic substance was also set up. The steps were as follows: Acquiring superant from the culture medium by centrifugation, discarding waste residue by filtration, concentrating by hollow-fiber membrane, treating with salting-out, dialyzing the product of salting-out, treating the dialysate by vaccum freeze dryer, dissolving solid , using the method of gel filtration chromatography with Sephadex G-100/G-75, and proof-test by filter-paper chromatography ,then preparation by thin-layer chromatography was carried out.
