A novel macrolide antibiotic, Maituolaimycin, was isolated and purified from the fermentation broth of strain Streptomyces regensis 099. Its novel structure based on spectroscopic analysis was determined, molecular weight was 600.03, and molecular formula was identified as C32H44N2O9. The preparative route was determined too.Detection and analysis method of Maituolaimycin was established, including biological detection method, thin-layer chromatography method and analysis high-performance liquid chromatography method. These methods can qualitative or quantitative detect and analysis various matrix samples.Process conditions of isolating Maituolaimycin from fermentation broth by macroporous adsorbent resin were determined. NKA nonpolar resin was screened as the best adsorbent, and adsorbent conditions were optimized: the pH was maintained at 10.32, flow rate of adsorption was 1ml/min, 0.6% NaCl was feasible, and adsorbed maituolaimycin at resin NKA was 33.7mg/g resin. The desorption conditions were optimized: 20% ethanol was used for removing some impurity and pigment, 100% ethanol was used for eluting Maituolaimycin, and 100% ethanol with 1% HCl was used for regenerating resin. Desorption rate was 92.7%.Preparative HPLC conditions of Maituolaimycin were determined. And through preparative HPLC , Maituolaimycin pure sample was get, which degree of purity was 95%.
