Huaijiao, a Chinese tradtional medicine(CTM), is the dried and ripe fruit of the Sophorajaponical plant. According to the pharmacopoeia, it can relieve inflammation or internal heat and cool blood or stanch bleeding owing to its cold nature and bitter taste. It is therefore applied for treatment of liver disease and headache. Genistein or 4', 5, 7 -trihydroxy isoflavone, the chief activated composition of Huaijiao, can kill various cancer cells but protect the normal cells. Hence it has become one of the focuses of medicine research and development. Nevertheless, as Genistein has the framework of isoflavone, its medical application is much restricted because of its poor water solubility and fat-solubility. This thesis, with Huaijiao as the ingredient, offers a systemetic research on extraction, analysis and modification of Genistein. The thesis, consisting of four chapters, presents the author's scientific achievements from the following three aspects:1 . With the application of a modern extraction method - ultrasonic extraction, Genistein, the anticancer activated composition, is obtained from Huaijiao. The influence of the varieties, concentration, dosage, extraction time, and degree of the solvent on the efficiency of extraction are examined. The influence of concentration, dosage, and the circumfluence time of the hydrochloric acid on the efficiency of the hydrolysis are investigated. The most ideal extraction and separation technical process and parameters of Genistein are furtherly perfected and the new technolgy of the ultrsonic extraction of Genistein in Huaijiao is successfully established.2. Three new derivatives are obtained with the chemical modification of Genistein. The new compounds are 3'-sodium sulfonate-4', 5, 7-trihydroxy isoflavone, 3', 6-disodium sulfonate-4', 5, 7-trihydroxy isoflavone and 3'-sodium sulfonate-7-methoxy-4', 5, -dimethoxy isoflavone. The structures of these compounds are identified by the methods of IR and NMR.3. The RP-HPLC methods are applied for analysis of genistin, baicalin, genistein and baicalein in Huaijiao pills, Zhitejia pills and Radix Scutellariae. The result of HPLC analysis of genistein andbaicalin in huaijiao pills shows: The linear range for genistein and baicalin was 0.1~1 g and 0.3~4 g respectively, the average recoveries of genistein were 99.66%. 100.3% and 99.97% with corresponding RSD of 0.57%, 0.25% and 0.43%, the average recoveries of baicalin were 99.87%, 100.4% and 99.98% with corresponding RSD of 0.32%, 0.19% and 0.34%; The result of RP-HPLC analysis of genistein, genistin and baicalin in zhitejia pills shows that the linear range for genistein was 0.0 1~ 0.07 ug respectively, the average recoveries of genistein were 98.97%, 10 1.4% and 99.56% with corresponding RSD of 0.52%,0.86% and 0.75%. The linear range for genistin was 0.07- 0.4 g respectively, the average recoveries of genistin were 98.37%,105.23% and 102.4% with corresponding RSD of1.01%,0.96% and 0.89%. The linear range for baicalin was 0.06~0.4 g respectively, the average recoveries of baicalin were 100.5%, 103.0% and 99.78% with corresponding RSD of 0.78%,0.98% and 0.56%. The result of HPLC analysis of baicalin and baicalein in radix scutellariae shows that the contient of baicalin and baicalein in radix scutellariae that is obstained by tissue culture under the condition of diffirent pH changes regularly. When pH value is equal to 5.8, the contient of baicalin and baicalein reach maximum. The studies may provide a scientific basis for the quality control of Huaijiao pills, Zhitejia pills and optimal condition of tissue culture of radix scutellariae.
