Cervical vertebra disc is the familiar diseases of middle and old age people. The high rate is Vertebraterial cervical vertebra disc, and the dizziness is main symptom. Few of Chinese traditional medicine can cure it, so the Dingyunning granules is studied to cure it. The granules is composed of 10 materials such as Pueraria Lobata, Gastrodia elata, Ramulus Uncaria rhynchophylla etc. It can remove convulsion, suppress liver, melt stases, unobstruct venation, remove phlegm, nourish liver and kidney, release dizziness and sober. The granules is used for vertebraterial cervical vertebra disc. Having observed the 60 clinic tests, it's efficiency reach 96.7%.According to the property of the effective composition in crude herb, the extraction technology is determined. Gastrodia elata and Uncaria rhynchophylla are seeped with 65% alcohol, and solution (I) is acquired. The residue is decocted with Pueraria labata, Salvia mitiorrhiza, Paeomia lactiflora, Pimellia ternate, Bombyx mori, Polygonum multiflorum, Poria cocos (8,6,6 times) three times ( 2,1,1 h) . Deposit with 65% alcohol, the precipitate is desert. Imbibe the top solution (II) . Using orthogonal test for extraction technology. Mixing solution (I) and solution (II), decocting, drying, then mixing Buthus martensii Karsch powder and honey element .Then the Dingyunning Granules is made.Using three methods to determine Dingyunning granules. First, determined the content of gastrodin and puerarin with HPLC .The good condition for gastrodin is used Hypersil ODS (4.6 X 250mm, 5um) column, mobile phase is consisted of methanol: water (4:96) (adjusted to pH 4.4 using acetic acid) .The detective wavelength is 220nm and IR is . 15.48min. The good condition for puerarin is used Symmetry C18 (3.9 X 150mm, 5um) column, mobile phase is consisted of methanol: water (25:75) (adjusted to pH 4.2 using acetic acid) . The detective wavelength is 250nm, and tR is 13.22min. Second, thin layer chromatography (TLC) is used for Paeonia lactiflora, Polygomun multiflorum, Salviamitiorrhiza and Poria cocos. Last, Attempted to determine the fingerprint chromatography with HPLC, received two fingerprint chromatographys. As the standard to compare with the chromatography of three batch granules. The result shows the chromatography can analyze Paeonia lactiflora, Gastradia, Pueraria labata, Ucaria rhynchophylla and Salvia mitiorrhiza from the Dingyunning granules.
