For the sake of exploring new method, which may extend the use of thin-layer scanning chromatography in China, according to the digital image processing theory, a software for utilizing office-use scanners in qualitative and quantitative analysis of thin-layer chromatography was developed. The most optimizing software can transfer a series of color spots in a matrix scanned simultaneously into a peak-form graph, in which Y-coordinate represents the grayscale or color integral value of spots and X-coordinate represents the positions of spots. The relationships between the spots' grayscale or color integral value of six colors: basic fuchsin, fruit green, naphthol green B, KFe[Fe(CN)6], K2CuFe(CN)6 and tartrazine spotted on silicagel thin-layers, filter papers and printer papers with different concentration, volume and area were investigated. And the arnino acid samples are successfully analyzed with planer chromatography.Investigation show that the spots' grayscale or color integral value is proportional to their volume or concentration of the color solution or the total amount of a color compound of the solution. RSD are in the range of 1%~9%.The different sample medium and the different colors of the sample will affect the sensibility of the quantitative analysis. Toward those spots which are very small and narrow, increasing the scanning differentiate rate will increasing RSD. Theoretically all the sample which can be measured by visible light spectrophotometric method can be analyzed by this method. Using this method aspectrophotometric method can be analyzed by this method. Using this method a lot of colored samples can be spoted as a plane matrix, and the datum will be calculated by computer. This will improve the speed of analysis, decrease the workload and increase the relativity.This study show the amino sample can be separated by planar chromatography. And the error of quantitative analysis is less than 11%. The application of office-use scanner in quantitative analysis of thin-layer chromatography is possible.Comparing with the classical thin-layer scanning method, this method is cheaper, faster and more ordinary. This method not only can be used in thin-layer chromatography, but also can be used in trace and array nalysis. This method will make a new way for quantitative analysis in thin-layer chromatography.
