From Edgeworthia Chrysantha Lindl, a traditional herb of China, thirteen compounds were isolated by silica gel column chromatography and structures of eleven of them were identified on the basis of spectral data and chemical evidence. Their structures were determined as 4-(3-hydroxy-l-propenyl)-2, 6-dimethoxyphenyi-$ -D-Glucopyranoside(eleutheroside B, ilexanthin A. magnolenin. syringin I ). 8-(7-hydroxycoumarin-8-yl)-7-α -L-rhamnopyranosyl-coumarin(edgeworoside C II ). 7-hydroxycoumarin (Umbelliferone III ), 5,7-dimethoxycoumarin(limettin IV ). Dulcitol( V ), lH-iso-Indole-l,3(2H)-dione(0-Phthalimide VI ). 3-(7-coumarinyloxy)-6-methoxyl-7-hydroxycoumarin(Daphnoretin VII), β -sitosterol ( VI ), 3( β )-stigmast-5-en-3- 3 -D-Glucopyranoside(Daucosterol IX ) . 3-(7-coumarinyloxy)-8-(7-hydroxycoumarin-8-yl)-7-hydroxycoumarin rhamnoside (Edgeworoside A X) and Sucrose( XI). Compound I , V, IX and XI were isolated from barks and stems of this plant for the first time and compound VI and VI were isolated from flowers of this plant for the first time, too. It is essential to point out that compound I is a valuable chemical constituent that has various bioactive effects.A simple, fast, sensitive and accurate RP-HPLC method for separation and determination of compound I in barks and stems of Edgeworthia Chrysantha Lindl was also described with optimum mobile phase in this paper.Another part of this paper is mainly discussed about preparative isolation and, purification of compound I and compound II from Edgeworthia chrysantha lindl by high-speed counter-current chromatography (HSCCC). A two-phase system composed of ethyl acetate-ethanol-water (15:1:15, v/v/v) was selected by thin-layer chromatography (TLC). HSCCC is considered as a suitable alternative for the separation of glycosides due to its unique advantages, especially some glycosides difficult to isolate by the other chromatography. HSCCC is an effective complementary way to other conventional methods for isolation of chemical constituents from traditional Chinese herbs.
