Garlic products with high content of alliin and alliin with high purity had extremely high commercial values because of its unique pharmacology activation, but the exact detection and high-efficient purification separation process of alliin was restrained by catalytic effects of alliinase on alliin.The thesis aimed at easily catabolism of alliin because of the effect of alliinase studied the interaction between alliinase and alliin under many kind of conditions, result showed the proper protection of alliin was to inactivate the alliinase in Situ by blanching. The proper process conditions were listed as follows: the blanching medium was water, the blanching temperature was 80癈 and the blanching time was 20 min.After the alliinase was inactivated, the sample was purified by means of Supelclean LC-C18 solid-phase extraction. The Inactivation of Alliinase in Situ-Solid-Phase Extraction-HPLO method which could detected the content of alliin in garlic was set up. The chromatographic conditions were listed as follows: TMS column was used; the detection wavelength was 214nm; the mobile phase was water; The flow rate was 0.4 ml/min. Under this chromatographic conditions, the RSD was below 1.33% when n is six, the recovery of standard addition was 96.0%~99.5%, and no effects of alliinase on the detection of the content of alliin compared with OPA, thus the content of alliin could be more accurate detected.Finally, the extraction process of alliin in garlic was particularly studied, and obtained the optimal process parameters.1 The optimal conditions for extraction of alliin was achieved when the garlic was mixed with 60% ethanol of the extration solution in the ratio of 1:5(g/mL) and extracted for two hours at 50C .The content of alliin was 1.75 % and extraction rate 4.68 mg/g.2, Crude Extracts was purified by means of the Alcohol-Sedimentation method, the better results could be obtained when the volume of the extraction solution and ethanol in the ratio of 1/8, and alliin content of the extract was 5.57% , the extraction rate of 9.67mg/g could be obtained.3 Alliin could be purified by 732 type cation exchange resin, the optimal process parameters might be achieved when the pH of the original solution was 2, the concentration of the original solution was 5.57mg/mL, the flow rate of the original solution was 1.0BV/h; The eluent was 1mol/L aqueous ammonia and the velocity of flow was 0.5 BV/h. After purified, the content of alliin in the extracts was about 40%, the recovery rate was approximately 60%, and the fine alliin could be obtained by means of recrystallization.
