Soyasaponin is one of the important components in soybean, which has some important bioactivities and physiological activities, such as reducing plasma cholesterol and guarding against atherosclerosis. It has great economic potential based on its medical functions as well as industrial functions, such as vesicant, emulsifier, stabilizer and flavor additive. In this thesis, the procedures of ethanol aqueous solution extraction were optimized by single factor experiment; A purification method was established with combination of AB-8 macroporous absorbent resin and cation exchange resin chromatography; A high-purity soyasaponin compound was obtained by preparative HPLC; A colorimetric method based on their aglycone or soyasapogenol for quantitative analysis of soyasaponin was established. The main results are shown as follows:At first, an opitimized method for extracting total saponins from soybean germs was investigated, using ultra sonic pretreatment in ethanol aqueous solution before macroporous absorbent resin extraction The optimum condition was as following: the marterial was defatted for 3 hours by petroleum ether, after then, it was extracted with 70 % (v/v) ethanol aqueous solution for 4 hours assisted by ultra sonic treatment for 45 mins at 20 KHz, and the solution to elute soyasaponins from macroporous absorbent resin was...
