This thesis has studied the analytical methods of determination of Functional Components of grape seeds.six new determination methods of Oligomeric procyanidins are reported. The methods have been applied to assay the samples and satisfactory results were obtained. It includes seven parts as follows:Part 1: Review. A review of the published analytical methods for determination of Oligomeric procyanidins is given.Part 2: Indirect method of Atomic Absorption Spectroscopy to Determination of Oligomeric procyanidins.A new method of determining Oligomeric procyanidins by atomic absorption spectroscopy was studied. The method is based on the reaction oligomeric procyanidins with Cu2+ and lead to yellow precipitate.The precipitate is centrifuged to subside. The Cu2+ in supernatant is determined by AAS,and the concentration of oligomeric procyanidins is obtained. The linear region was 3.0~30.0 u g/mL RSD=1.2 % and the recovery is 98.9-102.0%. when the method is applied to determine oligomeric procyanidins, It can obtain good result.Part 3: Determination of Oligomeric procyanidins by Ammonium molybdate Spectrophotometry.A new method for determining Oligomeric procyanidins by ammonium molybdate spectrophotometry was studied. The basis is Oligomeric procyanidins could react with ammonium molybdate and the yellow ester molybdate would be presented in sour solution. The linear region was 5.0-110.0 u g/mL the recovery is 97.8-102.5%,When the method is applied to determine oligomeric procyanidins,It can obtain good result.Part 4: Determination of Functional Components in Grape Seeds by Ferric Ammonium Alum-Potassium Ferricyandis.A new method of determining oligomeric procyanidins by ferric ammonium alum-potassium ferricyanide was studied. This basis is oligomeric procyanidins could react with ferric ammonium alum-potassium ferricyanide and the blue complex would be presented in acid solution, The linear region was 0.1 ?20.0 u g/mL, RSD=1.1%, and the recovery is 95.0~103.2%, when the method is appliedto determine oligomeric procyanidins, It can obtain good result.Part 5: Determination of Oligomeric procyanidins in Grape Seeds Using Flow-injection Inhibitochemiluminesce. A novel flow injection methodasith inhibitory chemiluminescence system for the determination of Oligomeric procyanidins was presented. Oligomeric procyanidins could inhibit the CL reaction of Luminol- C1O-system could be siminitized in the presence of Oligomeric Procyanidins. The increase of CL emission was correlated with Oligomeric Procyanidins concentration in the range from 0.8 to 24.0ug/mL, the detection limit was 0.3ug/mL and the relative standard deviation was 1.2%, The method was applied to determination of leucoge in pharmaceutical preparations with recovery of 98.0% and 102.5 %.Part 6: Determination of Functional Components in Grape Seeds by Ammonium Cerium Sulfate. A new method of determining oligomeric procyanidins by ammonium cerium(VI) sulfate was studied. The method is based on the reaction in strong acid solution of oligomeric procyanidins with Ce4+ ,The amount of residual Ce4+can be determined by spectrometry,Then the content of oligomeric procyanidins can be determined indirectly, The linear region was 0.12-10.0 u g/mL RSD=l.l%and the recovery is 97.2~102.8% with a detection limit of 0.04 u g/mL. when the method is applied to determine oligomeric procyanidins, It can obtain good result.Part 7: Determination of Oligomeric Procyanidins in Grape Seeds Using Flow Injection- Chemiluminescence Inhibition. A novel flow injection methodasith inhibitory chemiluminescence system for the determination of Oligomeric procyanidins was presented. Oligomeric procyanidins could inhibit the CL reaction of Luminol-H2O2-KIO4-system could be siminitized in the presence of Oligomeric Procyanidins. The increase of CL emission was correlated with Oligomeric Procyanidins concentration in the range from 0.4 to 20.0ug/mL, the detection limit was 0.06ug/mL and the relative standard deviation was 1.4%, The method was applied to determination of leucoge in pha...
