Ganoderic Acids' extraction of which by former researchers were all from fruit bodies, spores, or mycelia gained by Ganoderma lucidum submerged fermentation.Our research prove ganoderic acids yield by pH controlled fermentation is superior to uncontrolled does. New technique improves yield of ganoderic acids 26.5%.The intracellular ganoderic acids of mycelia produced from submerged fermentation by Ganoderma lucidum were extracted by the following conditions: extraction temperature, 91.6℃; extraction time: 4h; and mycelia: water ratio: 1:2.4;which was optimized by RSA. The maximum yields of ganoderic acids was up to 0.291g/kg wet mycelia.The composition of elution fluid, sample amount, column specification and the flow velocity of the elution fluid for the separation and purification of ganoderic acids by chromatography on a silica gel column were studied. The feasible conditions for the separation of ganoderic acids were as follow-ing : crude ganoderic acids were were eluted stepwisely with CHCl3:CH3OH=90:10,95:5,98:2 three kinds of elution fluid compositions under the ratio1:45 of sample : silica gel, the column specification was 40×500mm and the velocity of the elution fluid was 0.9 ml·min-1.We obtained 5 ganoderic acids respectively named as S1,S2,S3,S4,S5 in the broth of Ganoderma lucidum submerged fermentation that were detected by silica gel thin-layer-chromatograhy and UV scanning methods. We did the IR spec-troscopic analysis, MS spectrum , 13CNMR and 1HNMR spectrum of S3, Which hada relative lager amount. The IR spectrum showed that S3 belongs to tetracyclic triterpenoid acid, the MS spectrum gained the molecular weight of S3 to be 476,the 13CNMR and 1HNMR spectrum proved that the molecular formula of S3 is C27H40O7 and its structure.It was concluded in our experiment that GLFB and ganoderic acids had the effects of relieving cough, facilitating expectoration and controlling chronic bronchitis.
