During the process of the modernization of nature medicine, supercritical fluid extraction is thought as a clean technology with high efficiency and has received much attention. This thesis mainly investigated two kinds of processes for extracting volatile ingredient from and for extracting diosgenin from D. Nipponica M. The first section of this paper is to explore extraction of volatile oils from Radix Aucklandiae with SC-CO2. Orthogonal test was designed in order to find the suitable parameters. The effects of extraction temperature, pressure and flux of CO2 on the yield of volatile oils were analyzed .the optimal conditions were obtained. The effects of temperature play the most important role. The extraction yield reached 3.69% under the conditions of the temperature of 35℃ and pressure ranges from 20 MPa to 25 MPa. The pilot study has been operated further based on the lab scale .Using dehydrocostuslactone as reference substance, the production of pilot study is analyzed by HLPC. As a result, the content of dehydrocostuslactone in volatile oil is more than 38%. The second part of this paper is to study the extraction of diosgenin from D. Nipponica Makino on the base of the prophase work. It aims to make use of ultrasound to retreat raw material. As a result,ultrasound don't have an idea effect on the extraction. The further experimental results displayed that the value of pH of hydrolization has an effect on the extraction yield of diosgenin. When no neutralizing, a max. yield of diosgenin is achieved. Diosgenin extraction yield has a sharpen decline along with incremental pH . When pH>4, pH have a weak effect on the extraction of diosgenin. The process of hydrolization and SC-CO2 extraction has been studied,too. The resoult showed the value of pH has an effect on the SC-CO2 extraction and the yield of diosgenin reaches the max. at pH=7. During this research, moisture is a key factor. To have an idea gathering, hydrolyzation shoul be dried.
