ShuShe was selected as the strain for biosynthesis of Ganoderma lucidum polysaccharides. This text research into GLP production, condition of extraction, separation and purification, and the measurement of the molecular weight in order to find an effective way leading to high yield of GLP, and the main research results were as follows:Six Ganoderma lucidum strain ShuShe were examined by liquid and solid fermentation for the GLP producing capacities. The results showed that the highest yield of GLP among six strains was ShuShe.The effects of carbon source, nitrogen source, salt and the optimum initial pH value of medium on GLP produced by ShuShe were studied in liquid and solid fermentation respectively. The liquid and solid fermentive medium of high yield was selected by orthogonal experiment. The liquid medium was as follows: 3% maltose; 0.4% Peptone; 0.15% KH2PO4; 0.05% MgSO4; the optimum initial pH value of medium was 6.0. And the solid medium was composed of 100% bran, 1.5% sucrose, 0.05%, KH2PO4, 0.1% MgSO4, and 140~170% water. The optimum initial pH value of medium was original.In liquid and solid fermentation, fermentation technology condition in their highest yield GLP culture medium was studied respectively. The liquid and solid fermentation technology cndition of high yield GLP were selected by orthogonal experiment. The optimum liquid fermentation technology condition: fermentation temperature 28℃, fermentation duration for 136h, and inoculated rate 5%. The optimum liquid fermentation technology condition: fermentation temperature 30℃, fermentation duration for 178h, inoculated rate 0.6%.The production strain was inoculated into the optimum liquid and solid fermentive medium, and fermented in the optimum liquid and solid fermentation condition respectively, and then examined the GLP yield. The result showed that GLP produced by the solid fermentation was 58.8 g/kg; therefore GLP produced by the liquid fermentation was 52.5 g/kg. The yield of GLP by the former method was higher than the latter . comparing with the liquid fermentation and the solid fermentation, the fermentation duration of the solid fermentation was shorter 90h. fermentation are complicated relatively, and need to be fermented in ferment bottle.the productions of the liquid fermentation are polluted easily in fermentation, the dry expenses are also higher,and the investments of the production are bigger.Although the duration of the solid fermentation is long, the technology and equipment are very simple,and the production isn't polluted easily,if mold is controled in common situation,pollution can be avoided.Furthermore,the dry expenses are lower,the investments of the production are smaller.Seen from the cost, the solid culture matter bran is the waster of the agricultural product,and attaining 58.8 g/ kg GLP production needn't the nitrogen source, the carbon source amount is also lower than the liquid fermentation,in addition,the equipments of the solid fermentation are simple,the dry expenses are also lower,so compared with the liquid fermentation,the product cost of the solid fermentation lower to a great extent.Therefore,according to this experiment, the solid fermentation for GLP production was feasible, and could be used as a new method of GLP produced from now on. The condition of extract was developed in this experiment, select the optimum condition of extract, the results was as follows: extract solvent was distilled water; the radio of substance and water was 10:1; extract temperature was 90℃; extract duration for 60min. The fermented products was extracted by the optimum extract condition, and the yield of GLP was increased 13.4%GLP1 and GLP2 were identified by separating the solid fermentation GLP, and then those two polysaccharides were purified respectively. The result of identify of purity of GLP showed that GLP1 and GLP2 were all homogeneous compositions, and was up to the qualitative standard and quantitative standard.
