Insulin was conjugated with water-soluble fibroin and sericin proteins by treatment with glutaraldehyde and the processing conditions for the bioconjugates were studied in detail. The study used enzyme-linked immunosorbent (ELISA) kit for the quantitative measurement of insulin. The recoveries of fibroin-insulin and sericin-insulin conjugates were 67.8% and 49.2%, respectively; Half-life in serum of the two insulin conjugates were 30.2h and 28.4h, respectively, which were nearly three times than normal insulin injection. And activities of the two insulin bioconjugates were investigated by the model of diabetic rats caused by alloxan nonohydrate. After subcutaneous injection of the two insulin conjugates (5U/Kg), blood sugar assay showed that the two insulin conjugates lasted nearly 24h. Furthermore, the two insulin conjugates were safe, innocuous, and had no immunity in rats and rabbits.
