Objective L-Asparaginase is one basic drug in the treatment of Acute Lymphoblastic Leukemia (ALL), but its half-life time is too short and it is easy to arouse allergic reaction, in this study, We used native water-soluble silk fibroin to modify L-asparaginase in order to overcome its usual shortcomings mentioned above.Method L-Asparaginase was conjugated with water-soluble fibroin by cross-linking agent-glutaraldehyde, furthermore, enzyme dynamics and immune characteristics in vivo of modified L-asparaginase were also studied.Results The recovery of modified L-asparaginase was high (76.05%). The stability of modified L-asparaginase also increased, and its half-life time (70 hours) was much longer than that of native L-asparaginase (30 hours). The ability to arouse allergic reaction of modified L-asparaginase (antibody titer: 21) was significantly low than that of native L-asparaginase (antibody titer: 24). The Km of modified L-asparaginase (Km(app) = 0.844><10"3 mol-L"1) was 5.6 times lower than that of native enzyme. The affinity of modified enzyme to substrate L-asparagine elevated.Conclusion Silk fibroin-L-asparaginase bioconjugate;could overcome the common;shortcomings of native L-asparaginase, which suggest that this modified enzyme is worth studying and exploring as a new chemical drug.
