This paper studied red kojic rice including the isolation of Monascus spp., screening by mutagen, submerged fermentation and separating Monascus pigment and Monacolin k from red kojic rice, the effects of anti-fatigue and regulating blood-lipid were also evaluated.I Screening a High-productive Monascus Pigment Strain andPreliminary Study on Its Submerged FermentationFive strains of Monascus spp (M1-M5) were isolated and screened from Hongqu products. Then 5 Hongqu samples were prepared with the 5 strains by submerged fermentation, a strain of Monascus spp (M3) which products high Monascus pigment was choosert to be original strain. Through the mutagen of 60Co- Y ,a mutant strain of Monascus spp (M3-2) from M3 was acquired, which can produce much higher Monascus pigment than M3. The total color value of the fermentative mycelia and fermentation broth were increased 27.08%, 25.90% respectively. And the quality of Monascus pigment gained by the liquid fermentation of M3-2 was also studied.II The extract and isolation of Monascus pigment and Monacolin k inred kojic riceIn this chapter, a new method, i.e. the combination columns with silica gel column and neutral aluminum column, is brought forward to use separating the bioactive components, red pigment, yellow pigment and Monacolin k from red kojic rice, and the color shade of Monascus red pigment and yellow pigment showed the separate effect was well. The basic character of the three components was studied and the content of Monacolin k were determined by HPLC in Red kojic rice before and after isolation.III Preliminary study on anti-fatigue effect of Monascus red pigmentFifty-six mice were divide into 4 test groups at random, i.e. 3 drug groups and 1 normal control group. After took Monascus red pigment at dose of 0, 50, 100 and 200 mg kg-1 d-1 respectively for 4 weeks, measured swimming time, liver glycogen and serum BUN for test mice. Compared with normal control group, swimming time and liver glycogen of the mice in drug groups were increased while serum BUN was decreased. The result showed that Monascus red pigment has anti-fatigue effect.IV Study on regulating blood-lipid of Monascus red pigmentAccording to serum lipids level, 48 female Kun-Ming mice were divided into 6 groups: normal control group; high-fat control group; Gynostemma pentaphyllum control group; Monascus red pigment low, middle, high dose group; normal control group was fed basic diet and the others groups were fed high-fat diet for six weeks. After taking Monascus red pigment for 6 weeks at the dose of 10mg kg-1 d-1, 50 mg kg-1d-1 and 100 mg kg-1 d-1, Blood lipid level of the experimental mice were determined after starved 12h. The results showed that TC, TG, LDL-C level in serum and TC, TG, MDA level in liver of all Monascus red pigment groups decreased significantly compared with high-fat group, while HDL-C level in serum and the activity of SOD in liver are higher than that of high-fat group; It was indicated that Monascus red pigment has effect on lipid metabolism.
