Zhiganning Capsule is a kind of TCM agent which can be used to cure fatty liver.On the basis of clinic application, the compound prescription of Zhiganning was verified to be potent in curing fatty liver. The extraction methodology of the prescription was established and optimized with modern advanced techniques, while the dosage form was changed from traditional decoction to capsule. In this way, some weaknesses such as low contents of effective components, inconvenience of taking and carrying were got rid of. Objective: To establish the extraction methodology for Zhiganning Capsule, so as to get over the drawbacks existing in decoction.Methods: â‘  All the herbs were divided into two groups: aromatic herbs and non-aromatic herbs. The former was distilled to get volatile oil; the latter was abstracted with alcohol and purified by water-precipitation. â‘¡ Extraction methodology of aromatic herbs was determined by testing the rate of water-absorption, amount of water-adding and extraction time; Orthogonal test was adopted to optimize the extraction methodology of non-aromatic herbs with three indexes (extract yield, contents of gentiopicroside and emodin )â‘¢ The content of gentiopicroside and emodin was determined by HPLC. â‘£ To test the stability and practicability of the extraction methodology 100-fold raw materials were put into production with quality control.Results: â‘  Volatile oil:10 times amount of water under reflux for 6 hours; non-volatile components: refluxing with 10-fold 95% alcohol for 2 times and each for 1 hour,the condensed extractive was mixed with water to precipitate the impurity at a concentration of 70% alcohol. â‘¡ The capsules were also qualified when produced in mass.Conclusion: A technological process of the capsules preparation, which is economical, reasonable, and suitable for industrial manufacture, was underwent. The residue of aromatic herbs from which volatile oil had been isolated by distillation and other non-aromatic herbs were combined and extracted with alcohol, the condensed extractive was mixed with water to precipitate the impurity at a concentration of 70% alcohol.
